Object:To estimate the intervention effect of yangxinhuoxuejiedu fang on the JAK/STAT signal in the myocardium,to investigate the possible mechanism that make yangxinhuoxuejiedu fang take effect,and to seek the new target spot in the treatment of VMC.Method:The healthy kunming mice of SPF level were fed with regular feed for one week,then inject coxsackievirus to establish the model of VMC.The VMC successful model mice were randomly divided into the VMC model control group,ribavirin group,TCM low dose group,TCM medium dose group,TCM high dose group,another mice were as the normal group.Normal group and VMC model control group were given normal saline,the TCM low medium high dose group were given yangxinhuoxuejiedu fang,ribavirin group were given ribavirin granules.After 14 days,observe the conditions of the mice:survival situation,weight changes,and pathologic changes of pancreas by means of HE staining.Detect the serum myocardial enzyme,and detect the level of IL-6、IL-10 by enzymelinked immune sorbent assay(ELISA).Observe the expression about FAS and FASL in the myocardium by immunohistochemical method(IHC)and JAK2、STAT3 protein content in the myocardium by westernblot.Result:1.General conditions of the mice:with high or medium dose of yangxin huoxue jiedufang,the living quality can be improved.The mice in TCM groups have a low death rate and large size,Among these,the mice in TCM medium groups are largest(p<0.05).2.Effection on myocardial enzyme:Compared with model control group,CK-MB was reduced in each treatment group(P<0.05)。CK-MB in TCM meduim dose group significantly reduced of model group(P<0.05).Compared with the mice in the blank group,each treatment group except of TCM medium dose group,LDH level was increased(P<0.05),compared with the model control group,LDH in each treatment group was reduced(P<0.05),LDH in TCM medium and high dose groups reduced of model group obviously.3.Pathological changes:Myocardial damage model control group is heavier,myocardial cells appeared first focal lesions,Then flake degeneration,necrosis,focal inflammatory cells distribution around the necrotic myocardium.Traditional Chinese medicine(TCM)middle and high dose group of HCM is lighter.4.Detection of ELISA expression in the model group,IL-6 of surum increased outstandingly,there was obviously discreoancy compared with the control group(P<0.05).Compared with the control group,each treatment group equality depressed the level of IL-6,there was significantly reduced in TCM medium dose group.Detection of ELISA expressin in the model group,surum IL-10 dncreased,there was obviously discrpancy compared with the blank group(P<0.05).Compared with the model group,the level of IL-10 in each treatment group,all increased,there was obviously incresed in TCM medium dose group(P<0.05).5.Immunohistochemical method compared with the blank group,model group mice the expression in cardiac muscle tissue significantly enhanced(P<0.05).The expression of FAS and FASL of TCM medium and high dose groups in cardiac muscle tissue was at least.6.WestemBlot detection showed that in each model group mice cardiac muscle tissue of JAK2 increased significantly,compared with the model group,the content of TCM high dose group was the least,level of JAK2 in each treatment group was lowerer than the model group(P<0.05),the content of TCM medium dose group was the highest.Compared with the blank group,the content of STAT3 increased significnt,compared with the model group,TCM medium and high dose group of STAT3 decreased most.Conclusion:1.With Yangxin huoxue jiedufang,the living quality can be improved.The mice in TCM groups have a low death rate and large size.2.Yangxin huoxue jiedu fang can alleviate pathological changes,reduce the level of serum myocardial enzyme,reduce the level of serum IL-6 and elevated the level of serum IL-10,Inhibition of myocardial inflammation,which were On myocardial protection.3.Yangxin huoxue jiedu fang can reduce the level of Protein expression of FAS、FASL in myocardial tissue,reduce the myocardial cell apoptosis;reduce the level of Protein expression of JAK2、STAT3 in myocardial tissue,reduce the myocardial cell injury. |
