| Purpose:The paper is to observe impacts of jiedutongluo extract on myocardial fibrosis-relevant cytokines of experimental diabetic rat and therefore,to discuss function mechanism of jiedutongluo extract on diabetic cardiomyopathy.Method:1 A total of health Wister male 60 rats were fed adaptively for one week,and randomly divided the normal group(n=8)and the modeling group(n=52).For the modeling group,intraperitoneal injection of 1%STZ(55mg/kg)was administered to prepare experimental DM rat models,and 72h after modeling,fasting blood-glucose(FBG)was measured with tail venous blood.If a rat with an FBG≥16.7mmol/L and urine glucose+++was regarded as a successful modeling case.A total of 35 rats modeled with STZ were fed with average diets for another 12 weeks and then divided into a diabetic cardiomyopathy(DCM)modeling group(n=12),jiedutongluo extract group(n=12)and an Enalapril group(n=11).The extract group and the Enalapril group were respectively administered with16.7g/kg/d of jiedutongluo extract solution and 1.5mg/kg/d of Enalapril via intragastric infusion,while the DCM group and the normal group were administered with distilled water of same volume in the same way.The intragastric infusion lasted for 6 weeks.General conditions of the rats were observed,weights were recorded on a monthly basis,and blood glucose was monitored.At the end of the experiment after 18 weeks,hemodynamic indexes measured by left ventricle intubation were recorded;full heart weight indexes(full heat weight/body weight,HW/BW)were calculated;biochemical process was adopted to determine blood glucose and fat metabolic index;and ELISA approach was used to examine rat serum AngII and serum laminin(LN).2 Observe pathological morphology and ultrastructure change of myocardial tissue under optical microscope.3 Immunohistochemical method was used to detect expressions of NF-kB,TLR4,Types I andⅢcollagens and TGF-β1;and Semiquantitative RT-PCR was used to detect mRNA expression of TLR4 and HGF.Result:1 Jiedutongluo extract improves general conditions and clinical symptoms,including polydipsia,polyphagia,polyuria and emaciation,in the experimental DCM rats.2 Jiedutongluo extract inhibits cardiomegaly in the experimental DCM rats,showing effects of improving cardiac functions and reducing angiotensin II and LN levels.3 Jiedutongluo extract reduces FBG,TC,TG,LDL-C,Hs-CRP and improves blood glucose and fat metabolism in the experimental DCM rats.4 Jiedutongluo extract improves pathological morphology and ultrastructure change of myocardial tissue in the experimental DCM rats,showing a protective function for heart.5 Jiedutongluo extract inhibits and intervenes in myocardial fibrosis of the DCM rats and reduces expressions of NF-kB,TLR4,Collgen I,CollgenⅢand TGF-?1 in myocardial tissue.6 Jiedutongluo extract inhibits and intervenes in TLR4 expression in myocardial tissue of experimental DCM rats and enhances HGF expression,showing a heart protection role.Conclusion:In the experimental DCM rats,jiedutongluo extract improves general conditions and clinical symptoms(including polydipsia,polyphagia,polyuria and emaciation),inhibits cardiomegaly(showing effects of improving cardiac functions and reducing angiotensin II and LN levels),reduces angiotensin II level,LN level,and FBG,TC,TG,LDL-C,Hs-CRP and reduces blood glucose and fat metabolism,improves pathological morphology and ultrastructure change of myocardial tissue,inhibits myocardial fibrosis,reduces expressions of NF-kB,TLR4,Coll I,CollⅢand TGF-?1 in myocardial tissue,inhibits TLR4 expression in myocardial tissue,and enhances HGF expression,showing a heart protection role. |
