| Rosa rugosa ’plena’ have ornamental,edible and medicinal value.It was likely attacked by insect pests in its production.Chemical control might pollute environment and threaten human healthy.In this research,the variations of physiological and biochemical indexes in R rugosa ’plena’ induced by different inducing factors including mechanical wounding,JA,MeJA,Z-jasmone and SA were studied,and the affects of induced resistance of R.rugosa’plena’ on the detoxifying enzymes and feeding in Monolepta hieroglyphica were analyzed,in order to provide theoretical basis for the research on plant induced resistance.The results were as follows:1.Variations of physiological and biochemical indexes in R.rugosa ’plena’ induced by mechanical wounding:(1)nutrients:The contents of soluble protein and total amino acid of R.rugosa ’plena’were all increased by varying degrees mechanical wounding inducement,and the contents of soluble sugar were lower than control after this inducing.The variations of contents of soluble protein and soluble sugar had certain relationship with the different degrees of mechanical wounding.The maximum value of soluble protein was raised with the increasing of degrees of mechanical wounding,but the minimum value of soluble sugar decreased as the increasing of degrees of mechanical wounding.(2)defensive enzyme:Defensive enzyme activities were increased by mechanical wounding inducing.Different degrees of mechanical wounding could r:ise the activities of PAL、PPO、POD.The trend of which is appeared roughly that increased at the first then returned to the control levels.The relationship between variations of defensive enzyme and the degree of wounding is that moderate wounding could induce higher defense enzyme levels of R.rugosa ’plena’,but the activities of defensive enzyme were not raised with the increasing of the degree of wounding.The activities of defensive enzyme in the plants induced by 3 hole wounding(Three holes on each leaves was made and 1/3 leaves was wounded)were higher than that in the plants induced by 4 hole wounding(Four holes was made on each leaves and 1/3 leaves was wounded)indicated that there might be a threshold of this mechanical wounding.Suitable mechanical wounding could induce the activities of defense enzymes to be increased,but when the damage degree exceeded a certain limit,it could not make activities of defensive enzyme to be persistently increased.(3)secondary metabolites:The contents of total phenol and tannin were all increased by varying degrees mechanical wounding inducing.The contents of total phenol and tannin were not raised with the increasing of the degree of wounding.The 2 hole and 3 hole wounding(which means that 2 or 3 holes was made on each leaves and 1/3 leaves was wounded)induced relatively higher total phenol and tannin contents.Since phenol is the important defensive substance,so it could be speculate that mechanical wounding might induced the defensive reaction of R.rugosa ’plena’.2.Variations of physiological and biochemical indexes in R.rugosa ’plena’ induced by JA,MeJA,Z-jasmone and SA:(1)nutrients:The contents of soluble protein were increased differently induced by 4 kinds of compounds.The soluble protein was increased within the firstly day after induced after inducing,and then showed fluctuating change.The average value of soluble protein in the measurement period were higher induced by 1mmol/L of JA,0.5mmol/L of MeJA,1mmol/L of MeJA,0.5mmol/L of Z-jasmon,1mmol/L of Z-jasmone,0.5mmol/L of SA and 1mmol/L of SA.The trend of variations of soluble sugar contents appeared firstly increasing then reducing trend after inducing by those 4 kinds of compounds.The trend of variations of soluble sugar contents was consistent after inducing by all concentrations of JA.The contents of soluble sugar induced by JA were lower than that in the control group in the determination period.Among them,1.5mmol/L of JA induced the relatively lower soluble sugar content.Throughout the testing period,the contents of soluble sugar induced by 0.5mmol/L of Z-jasmone were significantly lower than that in the control group.(2)defensive enzyme:The activities of PAL、PPO、POD in the leaves of R.rugosa’plena’ was increased differently after inducing by 4 kinds of compounds.Induced by 0.5,’0.5 and 1.0 m mol/L of JA respectively,the activities of PAL,PPO and POD reached their maximal level on the 5th,3rd and 7th day respectively.Induced by 1.0,0.5and 0.5mmol/L of MeJA respectively,the activities of PAL,PPO and POD reached their maximal level on the 5th,3rd and 5th day respectively.Induced by 1.0,1.0 and 0.5 mmol/L of Z-jasmone respectively,the activities of PAL,PPO and POD reached their maximal level on the 5th day respectively.Induced by 1.0,0.5 and 1.0 mmol/L of SA respectively,the activities of PAL,PPO and POD reached their maximal level on the 3rd,3rd and 5th day respectively.(3)secondary metabolites:The contents of total phenol were all increased after inducing by those 4 kinds of compounds.The higher contents of total phenol were induced by 0.5mmol/L of JA,0.5mmol/L of MeJA,lmmol/L of Z-jasmone and 0.5mmol/L of SA respectively.The trend of the contents of tannin induced by those 4 kinds of compounds appeared increasing firstly then reducing.In the 1st-5th days after inducing,the contents of tannin all increased after inducing by those 4 kinds of compounds,but the time of peak value appeared after inducing by different compounds was varied.3.The effects of induced resistance of R.rugosa ’plena’ on the detoxifying enzymes and feeding in Monolepta hieroglyphicaThe activities of detoxifying enzymes in Monolepta hieroglyphica were inhibited to various degrees after feeding the R.rugosa ’plena’ which induced by different factors.The inhibition effects of detoxifying enzymes were relatively obvious after feeding the R.rugosa’plena’ induced by JA and MeJA.Different inducing treatment of R rugosa ’plena’ could reduce the area of Monolepta hieroglyphica feeding,it showed that different inducing treatment had certain effect of insect-resistant.4.Evaluation of induced resistance to insects of R.rugosa ’plena’ by different treatments:the capacity of induced resistance to insects of R.rugosa ’plena’ followed the sequence;1mmol/L MeJA,1mmol/L JA,0.5mmol/L MeJA,0.5mmol/L JA,1mmol/L Z-jasmone,0.5mmol/L Z-jasmone,3 hole wounding,1mmol/LSA,2 hole wounding,0.5mmol/L SA. |
