Temperature change in a single cell would be regulated by the stimulus from external environment, and this change would further edit the internal biochemical events in the cell. It's very important to understand the relationship between the single cell's temperature change andthe intercellular biological processes via single cell temperature detection, so that we could catch some significance in life science.In order to procure the single cell temperature detection, first it gives a confirmation of the temperature change in a single cell. Then,the single cell temperature detection system (SCTDS)is organized and the system standardization is finished. Further, the temperature of the cells high expressed with missing in metastasis (MIM) protein is detected by SCTDS, and it gets the information of single cell temperature change in the stimulus of camptothecin (CPT) and paclitaxel (PTX). Finally,the SCTDS's single thermocouple probe mode is improved and the coupling thermocouple probes SCTDS is designed to detect the single cell temperature change in the background of the ambient temperature.There were some findings and results in this article. Firstly, a simple equation was established to estimate the energy/temperature change in a single cell. This equation composed of extracellular energy input and intracellular energy conversion. Besides, the favorable characteristic of the temperature detection devices would help to accomplish the single cell temperature detection.Secondly, the temperature detection Pt-W thermocouple probes were prepared. The tip angle of the probe and electro-characteristics were studied for its efficient usage. The SCTDS was assembled based on these probes, and the measurement error of the system was defined as the systems' precision. The temperature-voltage relationship of the system was considered as the system's sensitivity. The work in this part supported the application of SCTDS in the single cell temperature detection.Thirdly, temperature of the single cell high expressed with MIM protein was measured by SCTDS. The temperature was increased in the 293 A cells high expressed with MIM, stimulated by CPT, and the intracellular ATP level had risen at the same time. These results were not happened in the normal 293A cells. Dissimilarly, PTX caused the 293A cells, with or without MIM expression, temperature increasing and intracellular ATP level rising, transiently.Fourthly, coupling thermocouple probes SCTDS was researched and developed for environment-single cell temperature measurement, and we got the favorite temperature detection sensitivity and accuracy. The coupling thermocouple probes SCTDS was applied in RAW264.7 cell's temperature detection stimulated by TTX. The single cell temperature was mostly decreased under stimulus of TTX.From the works of this article, we conclude that the temperature change in a single cell could be estimated by a simple equation, and it could be detected by available methods. Micro/nano thermocouple probes, with specified shape and electro-characteristics, could be used as temperature measurement probes in SCTDS for single cell temperature detection. Using the SCTDS, we got drug susceptibility changing and intracellular energy metabolism changing in the cells high expressed with MIM protein, stimulated by CPT. Temperature and ATP association detection would help to provide new ideas and methods for diagnosis and treatment of the diseases. The coupling thermocouple probes SCTDS would help us to obtain the environment-single cell temperature changing. This function could be applied in detecting the physiological states of the cells under different conditions of existence.In summary, it is very important to develop the single cell temperature detection and detection methods for understanding the cells stress reaction and intracellular energy metabolism to the enviromental stimulus. It would help us to touch the biotical sense of a single cell or organism.
