MicroRNAs (miRNAs) have been shown the potential for cancer diagnosis. We investigated whether serum miRNAs expression pattern could discriminate between patients with and without gastric cancer.MethodsThis study was divided into three steps:(1) MiRNAs sequencing of serum samples from6gastric cancer patients,6precancerous diseases patients and6healthy controls;(2) MiRNA selection by real-time qRT-PCR on46serum samples from15gastric cancer patients,16precancerous diseases patients and15healthy controls;(3) qRT-PCR validation on an independent set of plasma from95gastric cancer patients,70healthy controls, and85patients with gastric precancerous diseases.ResultsMiRNA sequencing analysis showed that37miRNAs were up-regulated and seven miRNAs were down-regulated in the serum samples from patients with gastric cancer compared that without gastric cancer. Seven miRNAs were validated in the serum samples from30gastric cancer patients and30healthy controls. We found that serum miRNA-331-3p and miRNA-197were significantly differential expressed in gastric cancer patients (p<0.05). Further large-scale validation experiment confirmed that both miRNAs expressions in serum were significantly higher in gastric cancer patients compared with either healthy controls or patients with gastric precancerous diseases. However, only the expression of miRNA-197in plasma was significantly associated with tumor invasion and with lymph node metastasis and tumor, node, metastasis stage.
