Objective1. By observe the effect of Si Jun-zi Decoction on cardiac function of rats and expression of Myocardial Proteins after chronic heart failure to investigate the mechanism Si Jun-zi Decoction treatment of chronic heart failure.2. Through research the influence of Si Jun-zi decoction on expression of myocardial proteins after chronic heart failure in rats in order to provide some molecular biology evidence for "heart-spleen correlation" theory of Chinese traditional medicine.MethodsTo establish rat model of heart failure by partially banding abdominal aorta for8weeks. The heart failure rats were randomly divided into4groups:Si Jun-zi Decoction preconditioning group(High and low doses), model group, positive control group, furthermore,1Sham operation group was established, After1month, myocardial hypertrophy index, cardiac function were determined, and the myocardial cells and interstitial change were observed with Microscope after myocardial tissue HE staining. After protein of left ventricular tissue were extracted from rat model of chronic heart failure Sijunzitang high-dose group and model group,2-D electrophoresis(2-DE) was employed to separate proteins, and the protein spots expressed differentially were picked out and subjected to tryptic in-gel digest and matrix-assisted laser desorption/ionization time of flight mass spectrometry (MALDI-TOF) for identification.ResultsIn Si Jun-zi Decoction preconditioning group, the cardiac function was markedly improved, and the myocardial hypertrophy index was reduced markedly compared with the model group. Compare with model group, there are31proteins increased and28proteins decreased in the Si Jun-zi Decoction group,9proteins in each group which displayed significant changes (Volume≥1.5) were subjected to MALDI-TOF, and5proteins were identified as follows:Serine protease inhibitor A3N、Pdhx protein、Suclg2protein、Creatine kinase S-type of mitochondrial、myosin regulatory light chain2, and3proteins of them are relate to energy metabolism regulating,2proteins are relate to myocardial remodeling and systolic function.ConelusionThe cardiac function and ventricular remodeling are improved profoundly in heart failure rats treated with Si Jun-zi Decoction. And Si Jun-zi Decoction could regulate the expression of Myocardial Proteins after chronic heart failure in Rats, heart function of the rats was improved may be related to energy metabolism regulating, myocardial remodeling reducing and systolic function improving.
