Background Carcinogenesis of colorectal cancer is implicated with heterogeneity and interacts with amount of genes, but there is no consensus of pathogenesis and prognostic biomarker until now. The genetic polymorphisms of TGFβ1signal pathway have been reported be associated with the development of colorectal cancer and can be a prognostic factor.However, there is no large study conducted in Chinese population.Methods We detected the SNPs of TGFβ1signal pathway, including3SNPs on TGFRI and2SNPs on TGFβ1in508colorectal cancer patients and641healthy people in China. Case-control study was carried out to analyze the relationship between these SNPs and colorectal cancer susceptibility in Chinese population, and prognostic value of these SNPs were also tested in the CRC patients.Results Mutant type of rs1800469and rs1800470significantly increased the risk of colorectal cancer, the corresponding OR were1.493and1.424,95%CI were1.076-2.071and1.030-1.968respectively. Moreover, the patients carried the mutant type of rs1800469and rs1800470got poorer survival (HR:1.504and1.231).Conclusion This is the first study to detect the association of SNPs in TGFβ1signal pathway and colorectal cancer patients and healthy people of China, and the results show great correlation with susceptibility and prognosis.
