Objective:This topic is to determine the expression of cx40and Ca2+-ATP of atria muscle tissue in paroxysmal atrial fibrillation big rats treated by acupuncture preconditioning. And to show effectiveness of acupuncture treating atrial fibrillation by related parameters of atrial repolarization such as P-wave width and dispersion, QT interval of atrial repolarization, chages of ultrastructure etc. To elucidate the molecular biological and cardiac electrophysiological mechanism of acupuncture treating paroxysmal atrial fibrillation and offer new thinking.Methods:1. Healthy male Wistar big rats weighing200-250g were chosen to establish paroxysmal AF model by7-day-injecting CaCl2-Ach mixture.2. Randomly divided the big rats with typical paroxysmal AF in ecg into control group, PAF group, acupuncture preconditioning group and cordarone group.3. Intervention methods:Acupuncture preconditioning group:acupuncture on double Neiguan30min before injecting mixture, the needle remained30min for7days, once daily. Cordarone group:rats were gavaged cordarone [4.5mg/(kg·d)] for7days, twice daily. Control group:rats were injected saline [1ml/kg] via tail vein. Related indexes were determined on the8th day.4. The expression changing of cx40and Ca2+-ATP of atria muscle tissue were determined by Western-blot. Judging indexes were AF duration time, P-wave width and dispersion, QT interval of atrial repolarization in ecg and chages of ultrastructure.5. Statistics analysis:Normal test, homogeneity of variance and t test were used for statistics analysis. SPSS16.0was used for statistics analysis. P<0.05stands for stastiscal significance.Results:1. Typical AF ecg appeared in model rats for f wave and p wave disappearing. Sinus rhythm came back after transient AF, f and P wave appeared. There were stastiscal significance on AF duration in Model group and control group (P<0.05) and which meaned successful modeling from ecg.2. There were stastiscal significance on P-wave width and dispersion1,4and7days after injecting mixture compared with control group (P<0.01)3. Durations of AF in both acupuncture group and cordarone group were shorten7days after modeling, and the results were superior to the model group (P<0.01).4. P-wave width and dispersion in both acupuncture group and cordarone group were decreased7days after modeling, and the results were superior to the model group (P<0.01).5. QT interval in both acupuncture group and cordarone group were decreased7days after modeling, and the results were superior to the model group (P<0.01).6. Expression of Ca2+-ATP of atria muscle tissue in both acupuncture group and cordarone group were up-regulated and the results were superior to control group.7. Expression of cx40of atria muscle tissue in both acupuncture group and cordarone group were down-regulated and the results were superior to control group.Conclusion:1. The model of paroxysmal atrial fibrillation big rats is reliable, repeatable and electrical remodeling of atria muscle tissue was similar with patients with AF.2. Acupuncture preconditioning on Neiguan point can up-regulate the expression of Ca2+-ATP of atria muscle tissue and reduce cell calcium overload caused by AF.3. Acupuncture preconditioning on Neiguan point can regular atrial repolarization by changing P wave width and dispersion.4. Acupuncture preconditioning on Neiguan point can regular electrical conduction of atrial by down-regulating the expression of ex40of atria muscle tissue.5. Acupuncture preconditioning on Neiguan point can regular AF and improve electronical remodeling caused by AF.
