| Objective:To study the protective effects of ischemic postconditioning against ischemic reperfusion injury of skeletal muscle, the time of application and the relationship between ischemic postconditioning and ischemic preconditioning.Methods:According to different treatment methods on ischemia-reperfusion injury,48 Wistar rats were divided into 6 groups. Using a rat amputation-like model,Wistar rats underwent temporary amputation at the level of the femur, excluding the femoral vessels. By measuring MDA, MPO, the extent of skeletal muscle death and apoptosis, Bcl-2 and Bax, protective effects were observed of postconditioning and preconditioning, preconditioning combined with postconditioning, postconditioning on 5 minute after reperfusion, the protective effects compare with the ischemic reperfusion.Results:ischemic postconditioning (30 seconds,three cycles) could protect skeletal muscle against ischaemic reperfusion, it can reduce the MDA and MPO in the reperfused areas,but also reduce the extent of muscle death and apoptosis, in the reperfused area anti-apoptosis factor Bcl-2 increase,Bax do not change.10 minutes of preconditioning can reduce the MDA and MPO in the reperfused areas, but the level of MDA and MPO is higher than postconditioning preconditioning also reduce the extent of muscle death and apoptosis,the protective effects is similar to postconditioning. Preconditioning combined with postconditioning can reduce the MDA, MPO and he extent of muscle death and apoptosis,but it is similar to the preconditioning or postconditioning alone. postconditioning on 5 minute after reperfusion can not reduce the MDA, MPO and he extent of muscle death and apoptosis in the reperfused areas.Conclusion:Ischemic postconditioning at the beginning of reperfusion can protect skeletal muscle against ischemic reperfusion injury, postconditioning on 5 minute after reperfusion lose protective effects. Preconditioning also protect skeletal muscle against ischemic reperfusion injury, but preconditioning combined with postconditioning don't offer additional benefit over preconditioning or postconditioning alone. |
