Purpose:To clarify the effects and its mechanisms of YiTangKang compound recipe treating diabetes mellitus(DM) on inflammation and macrobascular complication in DM rats so as to provide the theory base to apply Traditional Chinese Medicine in clinic.Material and method:Copied DM model with low grade inflammation and macrovascular complication by injecting STZ and high fat diet.Animals were randomized to normal Wistar control,model,YiTangKang groups and administrated water and YiTangKang compound for 4 weeks.In the experiment periods,observed general status and measured the amount of water drinking,food intake and body weight.And the end,C-reactive protein(CPR),monocyte chemoattractant protein MCP-1 and adiponectin by ELISA.The mRNA expression of peroxisome proliferator activated receptor PPARr and adiponectin in white adipose tissue and PPARr,AdipoR1,MCP-1 and nuclear factor NF-kBp65 in aorta by PCR,NF-kB activation using immuno histochemistry staining and observed thoracic aorta pathomorphology under light microscope.Results:The general status,the amount of food intake in YiTangKang groups were no significance compared with model group,but at the end of experiment,the body weight was gained in YiTangKang group campared with model group(P<0.01), concentrations of CRP and MCP-1 were all decreased in YiTangKang group compared with model group(P<0.01,P<0.05).The serum levels of MCP-1 were decreased in YiTangKang group was no significance compared with model group.YiTangKang group down regulated the mRNA expression of MCP-1,and inhibited the expression of NF-kBp65 and activation of NF-kB compared with model group in aorta,up regulated the m RNA expression of PPARr and adiponectin in white fat and PPARr in aorta,increased adiponectin concentration in serum compared with model group and decreased the amount of infiltrated cells and relieve lipid deposition in thoracic aorta.Conclusions:YiTangKang compound recipe had the role of anti-inflammation and preventing macrovascular complication in DM rats and which may be realized through these links as follows:YirangKang compound recipe inhibited the expression and activation of NF-kB in aorta,decreased the serum level of MCP-1 and CRP,up regulated the mRNA expression of PPARr in white fat and aorta and adiponectin in white fat,and increased adiponectin protein level in serum.
