[Objective]:We aimed at three points in the research.Firstly,it was to investigate the regularity for the change of PBMC mtDNA content and its association with HIV-LD in the HAART therapy.The second one was to find whether the change of mtDNA content could be a predictor for HIV-LD.The last was to see the impact of NRTI in different concentrations to the mtDNA content of preadipocyte culture in vitro and its association with LD.[Methods]:We established a method to quantify mtDNA content using real-time PCR. We chose the cryopreserved PBMC at baseline,month 6,month 24 of therapy,from patients regularly visiting our clinic for a long time and 3T3-L1 preadipocyte treated by d4T and AZT in different concentration,then total DNA was extracted,and mtDNA content was quantified.[Results]:The HIV/AIDS patients had higher PBMC mtDNA content than the normal comparison at the therapy baseline(p<0.01).The mtDNA content in the HIV-LD group was lower than that in the NLD group at each time point of therapy.In the HIV-LD group the half-year and 2-year PBMC mtDNA content was evidently lower than the base line(both p<0.01),and the changing of mtDNA content(appeared as soon as half an year) was earlier than the appearing of clinical HIV-LD(appeared one year later).In the NLD group,the PBMC mtDNA content didn't have a significant change after the treatment(p=0.161).mtDNA content decreased significantly in d4T containing regimen group after treatment(p<0.01),but the mtDNA content didn't change significantly in AZT containing regimen group after treatment(p=0.316).d4T and AZT could cause the cultured preadipocyte quantity and mtDNA content decrease, while the degree of the cell quantity and mtDNA content decrease had a conformity. d4T could cause a more severe decrease on mtDNA content and cell quantity,and it had a more significant association with the concentration.[Conclusions]:Our research indicated that:The decrease of PBMC mtDNA content during HAART therapy was associated with HIV-LD.The changing of PBMC mtDNA content was earlier than the appearing of the clinical HIV-LD,which could be one of the predictors of HIV-LD.The impact of d4T and AZT to preadipocyte's proliferation indicated that the decrease of mtDNA content caused by NRTI especially d4T played an important part in the HIV-LD process. |
