Distribution Of HIV-1 Infection Related Gene Polymorphisms In 20 Populations Of China

HIV/AIDS continues to spread globally and remains a worldwide pandemic.We notice that different regions have different epidemiology features.There are many factors attributing to it,including the function of immune system,the virulence of HIV-1 strain infected,the therapy after infection and genetic background of every individual.Futhermore,many studies support that the HIV-1 infection related gene polymorphisms are highly associated with the risk of HIV-1 infetion and the disease progression.Based on the two points,we hope to describe the polymorphism distribution of these gene locus in different populations,speculate the cause for it and intend to compare the susceptibility and disease progression among them.In the study,we screen the five HIV-1 related gene locus polymorphisms site in 20 Chinese populations(including 11 populations in Yunnan province) to study the distribution feature with PCR and PCR-RFLP and eveluate the relative risk for HIV-1 infection in every population.Polymorphisms of CCR5,CCR2,SDF,CX3CR1 and RANTES are reported to be involved in the HIV-1 infection or the development of disease progression to AIDS.These genes are followed.CCR5 is the coreceptor for the entry of HIV-1 virus,CCR2 and CX3CR1 are the minor coreceptors.RANTES and SDF are the nature ligands for CCR5 and CXCR4, respectively.After screening,we compute the relative hazard index in 20 different populations based on the different combinations of three gene locus genotype distribution(CCR5-CCR2-SDF) in every individual of the population.Our results suggest the things followed.(1) There is no individual with CCR5A32 mutation in our study group.The frequencies of CCR5A32 in 20 Chinese populations is dominantly lower than that of CCR5A32 in European populations.The frequencies of CCR2-64I,SDF1-3'A,RANTES In1.1C,CX3CR1-249I and CX3CR1-280M show the cline tendency from the south to north in China.(2)There is dominantly different distribution of CCR2-64I,SDF1-3'A,RANTES In1.1C,CX3CR1-249I and CX3CR1-280M in 20 Chinese populations.(3) The RH index in 20 populations are different,although the whole RH index are higher than that of European populations.(4)The populations in south have the relatively higher RH index and lower CX3CR1-249I-280M haplotytpe frequencies than that of populations in north. (5)The populations in the same language-speaking group share the similar CCR2-64I,SDF1-3'A,RANTES In1.1C,CX3CR1-249I and CX3CR1-280M distribution.(6)The popualtions geneticly releated or with the same origin also share the similar CCR2-64I,SDF1-3'A,RANTES In1.1C,CX3CR1-249I and CX3CR1-280M distribution.
