1.Effects of estradiol on the expression of FKBPs in the primary culture of rat hippocampal neurons.Objective:To evaluate the effects of estradiol and estradiol inhibitor ICI182,780 on the expression of FKBP12 and FKBP52 in the primary culture of rat hippocampal neurons.Methods:New-born SD rats were sacrificed and the hippocampi were dissected into small pieces and cultured in 24-well plates.MAP-2 immunocytochemical methods were used to identify hippocampal neurons.Neurons were divided into four groups: control,estrogen,estrogen+ICI182,780,and ICI182,780.The final concentration of estradiol and ICI182,780 was 10-8M.Using western blot we examined the expression of FKBPs protein and using rt-PCR we examined the mRNA of FKBPs.Results:At the ninth day of culture,there were over 92%of the cells that were verified as hippocampal neurons.There were both positive immunoreactivity of FKBP12 and FKBP52 protein and the immunoreactivity of FKBP 12 was up-regulated by estradiol treatment,as to the rt-PCR reaction we found the same results where FKBP 52 was not the case.Conclusion:There were both immunoreactivity of the two FKBPs in the primary culture of rat hippocampal neurons,and the immunoreactivity of FKBP 12 was up-regulated by estradiol treatment,as to the rt-PCR reaction we found the same results where FKBP 52 was not the case.2.Effects of estradiol on the cognitive function of male rats after lateral fluid percussion brain injury.Objective:To evaluate the effects of estradiol on the cognitive function of male SD rats after lateral fluid percussion brain injury.Methods:56 SD rats were divided into 3 groups which were control group,fluid percussion injury group and estradiol treatment group.Using lateral fluid percussion device the rats were injured.7 days after injury we used the Morris Water Maze to evaluate the reference memory and working memory of rats.Results:In the reference memory model estradiol treatment could not do any good on the memory of rats but in the working memory model we found it works.Conclusion:After lateral fluid percussion brain injury rats' cognitive ability were seriously damaged and estradiol treatment could improve the working memory while left reference memory fixed.3.Effects of estradiol on the expression of FKBPs in the rat hippocampus after lateral fluid percussion brain injury.Objective:To evaluate the effects of estradiol on the expression of FKBPs in the rat hippocampus after lateral fluid percussion brain injury.Methods:After the cognitive function assessment,rats were sacrificed and the hippocampi were dissected out.Western blot and rt-PCR were used to evaluate the FKBPs protein and mRNA.Results:Both FKBP12 and FKBP52 protein and mRNA were down-regulated after lateral fluid percussion brain injury.Treatment of estradiol could up-regulate the expression of FKBPs in the rat hippocampus.Conclusion:After lateral fluid percussion injury the expression of FKBPs were down-regulated in the ipsilateral hippocampus of male SD rats.Estradiol treatment could up-regulate the expression of FKBPs to normal level.
