| 1.ObjectiveThis study observed inhibition effect of modified prescription Liangfu (MPLF) on gastric cancer in vitro and vivo standing on TCM key pathogenesis of coagulated cold and blood stasis and previous researches about MPLF anticancer mechanism. The previous research on MPLF indicated it could have multifunctional anticancer mechanism including inhibit tumor growth, induce tumor cell apoptosis and suppress liver tumor angiogenesis. Making reference with tumor apoptosis research all over the world and the previous research on MPLF, this study explored anti-gastric-cancer targets of MPLF and elucidated its mechanism in several levels including tumor tissue ultrastructure, cellapoptosis and cancer-related gene expression in order to provide evidences for its extended application.2.MethodsIn vitro: After preparing different concentrations of MPLF- containing serum with,the inhibition on gastric tumor cell BGC-823 proliferation at different concentrations was observed for 48h,72h and 96h respectively by MTT. Cellular morphology changes are recorded by inverted microscope and transmission electron microscope (TEM) and gastric carcinoma BGC-823 cell apoptosis is assessed by flow cytometry.In vivo: After being established, implanted tumor models of human gastric carcinoma BGC-823 in the nude mice were intervened by MPLF in high, medium and low doses. Apoptosis-related factors (Survivin, Bcl-2, Bax, Fas, FasL and caspase-3) in these models were tested by immunohistochemical protocols so as to elucidate its anticancer molecular mechanism.3.ResultsThe results supported that MPLF has several functions as follows:â‘ different concentrations of MPLF-containing serum can inhibit gastric tumor cell growth and proliferation with dose-dependence;â‘¡c lassic apoptosis ultrastructure changes such as nuclear debris can be found under TEM in gastric tumors intervened by different concentrations of MPLF; the results tested by flow cytometry convinced that MPLF can induce human gastric carcinoma cell apoptosis;â‘¢MPLF high-dose group can inhibit implanted tumor growth of human gastric carcinoma BGC-823 in the nude mice (P<0.05);â‘£the mechanism of MPLF inducing gastric tumor cell apoptosis is probably due to down-regulate FasL, Bcl-2, Survivin protein expression and up-regulate Fas, Bax, Caspase protein expression.4.ConclusionsThis study indicated that MPLF can inhibit tumor cell growth both in vivo and vitro and induce tumor cellapoptosis, which can predict MPLF has a wide application prospect in cancer treatment. |
