Objective:To isolate Helicobacter pylori strains from clinical specimens,and establish two-dimensional electrophoresis(2-DE)maps of Helicobacter pylori in human gastritis and gastric cancer,and identify the differentially expressed proteins.Methods:The specimens of Helicobacter pylori strains in human gastritis and gastric cancer were clamped under gastroscope and isolated and identified with Gram staining and PCR test.The total proteins from Helicobacter pylori in human gastritis and gastric cancer were seperated by means of immobilized pH gradient-based 2-DE.The differentially expressed proteins were screened by using PDQuest analysis software, and were identified by peptide mass fingerprint(PMF)based on matrix-assisted laser desorption/ionization time of flight mass spectrometry(MALDI-TOF-MS)and database searching.Results:Helicobacter pylori strains were rucceffully isolated.A well-resolved and reproducible 2-DE pattern of Helicobacter pylori was obtained from patients with human gastritis and gastric cancer.14 of these proteins were identified.They take part in protein synthesis and degradation,chaperone,protection and detoxification,signal transduction,metabolism or cytoskeleton construction.Conclusions:Helicobacter pylori strains were successfully isolated from clinical specimens.A well-resolved and reproducible 2-DE pattern of Helicobacter pylori in human gastritis and gastric cancer is established, and fourteen differentially expressed proteins were characterized.
