Heat shock protein 27(Hsp27) is an important member of small heat shock proteinsub-family. Evidences based on the non-myogenesis cell culture studies have shownthat Hsp27 is a novel effective molecular in anti-oxidative stress and anti-apoptosis.The pilot study has shown that overexpreswsion of Hsp27 confers resistence to theapoptosis induced by hyperthermia (heat shock) in isolated mice heart, but themechanism is unclear. Furthermore, the role of Hsp27 in cardiomyocytes in vivo inintact animal still remains to be determined.ObjectiveTo determine the biological roles of Hsp27 in: (1) oxidative stress induced damagesin cardiomyocytes in vitro, (2) biochemical induced cardiac tocixity in intact mice invivo.Methods1. Studies in vitro —cloning and reconbination of human Hsp27 gene & to studythe effects of overexpression of Hsp27 on the oxidative damage in rat cardiac cellH9c21.1 Full length of Hsp27 cDNA obtained by RT-PCR was inserted into plasmidpcDNA3.1(+). The expression activity was tested by transient transfection with 293T.The stable transfected Hsp27 H9c2 cell lines (Hsp27 H9c2) were established byselection with G418.
