| In order to identify the role of genetic factors in pathogenesis of I°SS. We detected polymorphism of HLA-DRB1 (DR1-DR10), HLA-DRB3 (DR52), HLA-DRB4 (DR53) among 70 Chinese I°SS patients and 136 normal controls by using PCR-SSP (sequence specific primer) method. The patients and controls are living in the same geographic region. It was found that HLA-DR3, HLA-DR52 and HLA-DR2 allelic frequencies in the I°SS patients were significantly higher than that in normal controls. However, the allelic frequencies of HLA-DR5 and HLA-DR9 in patients with I°SS were much lower than that in control group. The results suggested that I°SS is positively associated with HLA-DR3, HLA-DR52 or HLA-DR2. In contrast I°SS is negatively associated with HLA-DR5 or HLA-DR9. We also found that the same genetic genes were shared among the siblings of I°SS family, which was consistent with the risk genes found in I°SS population. It was further suggested that HLA-DR3, HLA-DR52 and HLA-DR2 may be the risk genes of this disorder. Our studies did not demonstrate the relationship between HLA-DR3 or HLA-DR2 and anti-SSA or anti-SSB antibodies. However, We found that HLA-DR52 was associated with the anti-SSA and anti-SSB antibodies. Among the Chinese people, HLA-DR2(15) was the predominant allele of DR2 (87. 5%).
