| Interleukin-1(IL-1) is an important cytokine, which has many biological activities. IL-1 plays a key role in many pathological conditions, like inflammation, infection, fever, septic shock, auto-immune diseases and so on. It works through its high affinity interaction with IL-1 receptor, which is present on a wide variety of different cell types. There are two types of IL-1 receptors: type I rceptor(IL-1R â… ) and type â…¡ receptor(IL-1R â…¡ ).Soluble forms of IL-1R can be produced through protelytic cleavage of the extracellular portion of the two types of receptors.There are two sections in the project.In the first section, the metabolites of Actinomycetes 103 has been well studied. It was obtained from the IL-1R antagonist screening model targeting the recombinant mouse sIL-1R â… expressed from Sf9 insect cells.The fermentation supernate was extracted, isolated and purified according to the bioassay. 103A, as a kind of inhibitor of IL-1R, was obtained finally. It was elucidated as a kind of polysacchride through chemical analysis, SDS-PAGE and spectral analysis. The constitution of 103A was determined by capillary gas chromatography. The molecular weight of 103A was identified through gel chromatography method.In the second section, the metabolites of fungus 2205 has been well studied.
