| Objective This study was designed to investigate the mechanism that "horror hurt the pregnant rat" had effect on the neonatal rat brain development.Method ①The pregnant Sprague-Dawley rats aged 3 months were randomly divided into normal group, model group and bu Shen group(n=10 in each group).During gestational period, the rats in model group and bu Shen group were frightened by the cat four hours(2 hours Am,2 hours Pm) a day. The results were examined at postnatal day 1 and postnatal day 14.② The number, the data of growth and development of the neonatal rats were observed.③The cerebral cortex within hippocampus collected from each group neonatal rats at postnatal day 1 and postnatal day 14 were examined under light and transmission electron microscope. ④Blood samples were collected at the time of postnatal day 14 for the determination of free triiodothyronine(FT3)/free tetraiodothyronine(FT4) and thyroid stimulating hormone(TSH) were measured by using the chemiluminescence method. ⑤Agarose gel electrophoresis was used to dectect DNA fragmentation ,while the expression of Bcl-2/bax gene in the neonatal rats hippocampus was observed by the immunohistochemical method. ⑥ Activities of acetylcholinesterase and Choline acetyltransferase in the neonatal rats hippocampus at postnatal day 14 were determined by using UV-Vis spectrophotometer。 ⑦The expression of brain derived neurotrophic factor gene in the neonatal rats hippocampus of each group was measured by reverse transcriptase polymerase chain reaction.Result ①The number of neonatal rats in model group was less than in normal group(P<0.01),The growth and development of neonatal rats in model group was slower than in normal group ,while bu shen prescription can increase the low level. ②The morphology of the cerebral cortex within hippocampus in the three groups was observed: Under light electron microscope, some neurons were edema, the number of neurons showed decrease with a decreased number of dendritic arbors. Under transmission electron microscope, the abnormal structure of cell organ and karyopyknosis showed in nerve cell were observed in model group. While the normal and bu shen groups had no marked significance.③The blood serum level of FT4 in pups at postnatal day 14 of model group came forth lower than the normal group (P<0. 05) ,while bu shen prescription can adjust it into normal. The blood serum level of FT3, TSH in pups of the three groups had no marked statistical significance. ④A little DNA fragmentation was observed in model group,while it was none in normal group and in bu shen group.The expression of Bcl-2/bax gene in the neonatal rats hippocampus at postnatal day 14 of model group decreased as compared to in normal and bu shen groups. ⑤The activities of Ache and CHAT in the neonatal rats hippocampus at postnatal day 14 rised in model group as compared to in normal and... |
