Objective To study the CD26/DPP4 gene expression in ovarian carcinoma cell lines and its relationship with malignant behavior of ovarian carcinoma cells.Methods The expressive property of CD26/DPP4 gene among A2780, SKOV-3, HO-8910 and HO-8910PM cell lines were detected by techniques of DPP4 activity assay, cytometry and RT-PCR. The differences of the malignant behavior among the cell lines were examined by cell proliferating dynamical analysis, adhesive test, assay of incursion and chemotaxis. The correlation between CD26/DPP4 gene expression and the ovarian carcinoma cell lines activity of proliferation, adhesion, incursion and chemotaxis were analyzed, as well.Results The transcription of mRNA of A2780, SKOV-3, HO-8910 and HO-8910PM cell lines were 0.7512±0.0012, 0.5596±0.0015, 0.3369±0.0009 and 0.2777±0.0006, respectively. That of the A2780 cell line was significantly higher than the latter (P<0.001), and there were significant differences among the latter three cell lines (p<0.001).The protein expression of CD26/DPP4 gene were 657.83±1.14, 538.53±5.29, 130.50±1.46 and 33.14± 0.47, respectively. That of the A2780 cell line was higher than the latter three (p<0.001), that of the SKOV-3 cell line was higher than the latter two (p<0.001), and that of the HO-8910 washigher than HO-8910PM (P>0.001).The DPP4 enzyme activity were 0.79±0.02, 0.64±0.03, 0.21±0.02 and 0.18±0.01, respectively; those of the former two were significantly higher than the latter two (P<0.001), and there was no difference when compared the latter two cell lines (P=0.32).Among all cell lines, the proliferating ability of HO-8910PM was the fastest, while that of A2780 was the slowest (P<0.001).The counts of adherent cells among cell lines A2780, SKOV-3, HO-8910 and HO-8910PM were 42.0±2.0, 82.3±2.5, 113.0±3.0 and 138.0±3.0, respectively (P<0.001). Those of the latter three were higher than that of the first cell line; those of the latter two were higher than that of the second cell line (P<0.001). That of the cell line HO-8910PM was higher than that of HO-8910(P<0.001).While the counts in assay of incursive ability were 32.0±2.0, 48.7±3.1, 99.6±1.0 and 99.3±1.5, respectively. There were significant differences when compared the former two with the latter cell lines (P<0.001), while there was no difference between the latter two (P=0.881).The cell counts of migratory test were 35.0±1.0, 66.7±2.5, 103.3±3.1 and 104.0±2.6, respectively. There were significant differences when compared the former two with the latter cell lines (f<0.01), while there was no difference between the latter two (P=1.0), though more migratory cells in HO-8910PM cell lines.The correlation coefficients of the CD26/DPP4 gene transcription and the proliferating, adhesive, incursive and migratory ability of ovarian carcinoma cells were -0.962, -0.987,-0.975 and -0.991, respectively(f<0.001).The correlation coefficients of the CD26/DPP4 gene expression and those ability of cells were -0.886, -0.959, -0.988 and -0.962, respectively (f<0.001).The correlation coefficients of the CD26/DPP4 enzyme activity and those ability of cells were -0.900, -0.952, -0.997 and -0.978, respectively (f<0.001). Those of cell line A2780 were the highest, next to cell line SKOV-3. Whilethose of HO-8910 and HO-8910PM were the lowest.Conclusion There is a negative correlation when the expression, transcription of CD26/DPP4 gene and the activity of the enzyme were compared with the proliferating, adhesive, migratory and incursive ability of ovarian carcinoma cells. |
