The steroid-dependent asthma (steroid-dependent asthma, SDA) belongs to the difficult curing asthma category, is one of the difficult problems which the clinician faces. With understanding that the airway inflammation of asthma are essential, glucocorticoid become first-selected medicine for asthma treatment, and the using in a large amount of glucocorticoid not merely brings various kinds of poisonous side reactions , furthermore make the organism immunity drop, HPA axle function suppress seriously, cause glucocorticoid receptor (Glucocorticoid receptor, GCR) the competence or affinity drop at the same time, cause some patient reliance in GC finally, once did glucocorticoid reduce or stop using, namely cause the asthma to recur or aggravate ,the asthma of those patient form the steroid-dependent asthma. SDA have the airway inflammation exist continuously and HPA axle severed during dismissing from the medicine. The inflammation cause by T helper cell subsets of out-of-balance in proportion Th1/Th2, namely the function of Th1 (mainly secrete γ-interferon, IFN- γ) is suppressed , Th2 is in the state of activating, the latter secreting cytokine such as interleukin-4 (IF-4), interleukin- 5 (IF-5 ) which accelerate eosinophil (EOS), mastocyte, etc. to promote inflammatory cell differentiate, assemble and activate constantly and IgE synthesize, lead to the fact that responsivity of airway increase continuously, a large number of glucocorticoid medicine at the same time regulate competence of duplicating of GCRmRNA backward, make its quantity reduce or the activation is reduced, cause patient's reliance on the glucocorticoid. Glucocorticoid once reduce or stop using is it can cause the asthma to recur , even cause the death seriously. Therefore we must seek to the effective treatment project that can resume HPA axle function that suppress because the glucocorticoid is used for a long time in a large amount, can regulate Th1/Th2 ratio, reduce airway of high responsivity, reduce glucocorticoid treat consumption, lighten whole body from bad reaction, is it improve in an all-round way to patient quality to realize. In the opinions of my advisor professor Wu WeiPing, deficiency of yin and yang, stiring up of deficient wind, accumulation of phlegm and blood stasis in the body, simultaneous occurrence of cold and heat syndrome are essential pathogenesis of the steroid-dependent asthma(SDA) after use the glucocorticoid as the leading treatment medication. It held that the patients presented such pathologic change rules as from deficiency of yin and hyperactivity of evil fire before the steroid withdrawal to deficiency of yin and yang during the steroid withdrawal to deficiency of kidney-yang after the steroid withdrawal, with the syndrome of the stasis of sputum and blood throughout the whole process. Hereby she framed the three-step sequential therapy that compose by ZhiBaiDiHuang pill(smoothing bronchial wheezing by nourishing yin), WuMei pill(supplementing both the vital energy and blood, regulating cold and heat) and JinKuiShenQi pill(smoothing bronchial wheezing by nourishing yang). This way can help to dismantle steroid and control symptom of asthma. 1. Objective The purpose of this experiment is imitating SDA patient's pathology change ,so we inject dexamethasone on the venter of the model rats that have asthma. Through study the asthma model mouse that treated by glucocorticoid during the steroid withdrawal and after use traditional medicine and Western medicine the airway inflammation and HPA axle corresponding change situation, announce the evolvement rule and inherent material base for steroid-dependent asthma during the steroid withdrawal, probe into treat airway inflammation in asthma and resume HPA axle function mechanism of three-step sequential therapy. 2.Method 2.1 The function of lung is detected by the lung function instrument to minitype animal 2.2 Serum IFN-γ, IL-4,ACTH, steroid and lung tissue CRH were detected in the way of ezyme-linked immuneosorbent assay. 2.3 Lung tissue IFN-γ 和 IL-4 mRNA were...
