It is well known that the cognitive ability of the human being will be decreasedobviously over forty, which bring much trouble to lives. So, it is important for thecoming aging society to study on the cause of age-related cognitive ability decreaseand put forward the effective strategy. It is believed that the estrogen has theantioxidant effect and neurotrophic effect. It is found in the clinical data that themiddle-old-aged woman usually accompanies serious the learning and memoryimpairment when they experience the atrophy of the ovary, hypogonadism endocrinedisturbance and the menopause. The bushenyao can improve the productive functionand action of the wise-sharpen, although the mechanism is still unknown. Thearomatase is the limLited step of the synthesis of the estrogen, which exists in thebrain region related with the memory and learning beside sex gland. What kind of roledoes the aromatas play in the memory and learning? Whether the aromatase is relatedwith the effect of bushen and the wise-sharpen? They are all worthy to study further. 1 OBJECTIVES According to the hypothesis, after the concentration of the brain is kept at arather low level, the ability of learning and memory of the animal are detected by thebehavior study and electrophysiological method and pathological method. With the guide of the traditional Chinese medicine (TCM) theory, this essayscreen the fufang which has obvious wise-sharpen action and explore the action andthe mechanism of the prevention and cure of the middle-old-aged learning andmemory impairment. According to the bushen theory of the TCM, this essay explorethe bushenyishifang's action of the estrogen and aromatase in order to illuminate thiskind of fufang' effect on the age-related learning and memory impairment and itsmechanism. 2 METHODS and MATERIALS2.1 Establish the rat model with the low concentration of estrogen Three months old SD female-rats are randomLy divided into sham-operationgroup , ovariectomized (OVX) group , ovariectomized-Exmamtane-treated(OVX+EXE) group, except for the sham operation group, all the rat are removedtwin ovary. The OVX-Exmamtane-treated group ip 60mg·kg-1 exemantane once 3days.After 30 days, observe the ability of learning and memory by the step down, stepthrough, Morris water maze. The brains of half rats are obtained by the infusion ofparafonnaldehyde and fixed for the preparation for the paraffin sections andimmunohistochemistry of aromatase and hemetoxylin and Eosin stain.
