Palace Tumor Clear Serum Containing Experimental Research, The Mechanism Of Uterine Fibroids

Objective: Set up experimental model of human uterine leiomyoma cells/ smooth muscle cells in vitro to research the effect of steroid hormones(E2, P) and / or growth factor(IGF-K EGF) on proliferation of hysteromyoma cells/ smooth muscle cells . Observe the action of serum containing Gongliuqing on proliferation of hysteromyoma cells and stimulating factor which can improve proliferation of hysteromyoma cells based on serum pharmacological methods to probe into the mechanism of action of Gongliuqing capsule on hysteromyoma.Methods: Culture human isolated uterine leiomyoma cells/ smooth muscle cells and detect the optical density of uterine leiomyoma cells/ smooth muscle cells which are stimulated with subjects on 12h, 24h, 48hs 72h with the method of MTT.Results: 1. IGF, IGF+E, IGF+P, IGF+EGF had an action of promoting proliferation of hysteromyoma cells. IGF -, IGF+E2, IGF+P, IGF+E2+P, EGFs EGF+E2, EGF+P, EGF+E2+P, IGF+EGF possessed effect of promoting proliferation of uterine smooth muscle cells. 2. Serum containing Gongliuqing showed an action ofsuppression on proliferation of uterine leiomyoma cells. 3. Serum containing Gongliuqing could antagonize the effect of IGF , IGF+E2, IGF+P, IGF+EGF on proliferation of uterine leiomyoma cells .Conclusion: 1. Serum containing Gongliuqing showed a direct action of suppression on proliferation of hysteromyoma cells, which indicated hysteromyoma cells were target cells. 2. Serum containing Gongliuqing could antagonize the effect of IGF> IGF+E2, IGF+P, IGF+EGF on proliferation of uterine leiomyoma cells , which hinted Gongliuqing capsule acted on cell signal transfer pass of estrogeon/progesterone receptor and Growth factor receptor. 3.Suppressing proliferation of uterine leiomyoma cells and antagonizing the effect of IGF, IGF+E2, IGF+P, IGF+EGF was the mechanism of action of Gongliuqing capsule and indicated the function of improving blood circulation and removing blood stasis and resolving nodes and masses of that.
