Clinical Evaluation Of Kidney Treatment Mesangial Proliferative Glomerulonephritis Naoshuning Experimental Study Its Impact On The Mesangial Matrix Sclerosis And Related Factors

Purpose:Mesangial proliferative glomerulonephritis (MsPGN) is the most commonly type of chronic nephritis,Its main pathological change is mesangial cell proliferation and extracellular matrix expansion ,whatever the later is the key feature of glomerusclerosis .In this study ,to explore the clinical effect of ShenShuNing(SSN) on curing MsPGN and it抯 molecular mechanism in preventing mesangial matrix sclerosis from vivo and vitro.Method:(1)Choose 30 patients diagnosed as MsPGN after renopuncture who classified deficiency of Qi and Yin with deficiency of liver and renal,treated with SSN.To evaluated its clinical effect from the changes of main symptoms and biochemical pathology and immunology and blood coagulation mechanism.(2)To make the rat model of MsPGN by injecting steophylococcus enterotoxin B(0.4mglkg) into rat caudal vein at first and 7th day,feeding bovine serum albumin(2mgld), injecting Freunds adjuvant (2m1/kg) periodical (7th and 14th day) and treated with SSN,at the same time,use SheYanKangFuPian(SYKFP) as control. To observe the changes of pathologichistology and ultrastructure and the immune complex deposit in the MsPGN rat kidney tissue afer using SSN and SKYFP,and the expression of transforming growth factor- f3 1(TGF- f3 ~) and intercellular adhesion molecule-1(ICAM-l) and collagen IV(CoIIV) and fibronection(FN) in the MsPGN rat renal tissue by using Imrnunohistochemisty(SABC),ELISA and RT-PCR.(3)In the vitro, using different dose SSN pharmacological serum act on mesangial cell(MC) at different time under normal and angiotensin(AT II) condition, to observe MC proliferation and @ secretion of Col 1W. FN and its mRNA expression ~ secretion of TGF- P and its mRNA expression @~ secretion of ICAM-1 and its mRNA expression by using MTL ELISA.
