Alzheimer's disease (AD) which is a cause learning, memory and cognitive dysfunction in the central nervous system degenerative diseases, the incidence rate with age, aging increased significantly. Epidemiology, in recent years the incidence of AD was an increasing trend, and thus the method of prevention and treatment of this disease has become an urgent need. As the pathogenesis of AD is unclear, the pathogenesis of currently used drugs target, due to limited efficacy and short-term and less than ideal. Mentor Professor Gao Weibin use electric acupuncture treatment of senile dementia term clinical efficacy by years, this study by experimental analysis of its mechanism.Objective:According to the grouping studies of Alzheimer's disease in rats of the mechanism of electro-acupuncture of nuchal for treatment of senile dementia,to provide an objective basis for the clinical use of electro-acupuncture of nuchal in treating senile dementia.Methods:The Alzheimer's disease (AD) rats were divided into control group, sham injury group, model group, EA group(2W),EA group(4W), intraperitoneal injection of D-galactose and fed with aluminum chloride method of manufacturing damage model.Complex damage modeling, the rats appeared apathetic, less active, slow, loss of appetite, dull coat or hair removal,and the persistence of symptoms, which prompts modeling success.Treatment:select the bilateral "Fengchi" and "blood points" for acupuncture treatment. Use0.40mm×25mm acupuncture piercing3mm, followed by KWD-808Ⅱ pulse electro-acupuncture device, wires to connect the upper and lower needle handle, positive connection"Fengchi" negative connection "GongXue." The use of dilatational wave, frequency of4-20HZ, density cycle6S, strength in key muscles twitching as a slight degree, continued to stimulate20min, once a day, every needle6days, rest one day, Two EA-group were treated continuously for2weeks and4weeks. Project and detection methods:using the Morris water maze test, testing before making model, after building the model, therapy after a mean latency, number of average across the platform, and treatment behavior comparison; using HE staining and light microscopy were observed after treatment of rat hippocampal morphological changes were observed by electron microscopy; after the treatment of rats hippocampal tissue and cells within the ultrastructural changes; Using immunohistochemical methods to observe the treatment of hippocampal organization of the positive expression of the acetylcholinesterase (AchE), acetylcholine transferase (ChAT), dopamine receptor D1(DRD1) and serotonin (5-HT); in situ hybridization methods to observe the positive expression of CHATmRNA in hippocampal organization after treatment.Results:1.Behavior changes:(1) Mean escape latency Between the groups: by the EAON treatment, two of the rats in the treatment group,the mean escape latency compared with the model control group was significantly shorter, and there was significant difference (P<0.01), and the4weeks group was better than that of2weeks (P<0.01); Within the groups: model control group, two EAON group of rats in their group before and after modeling,the mean escape latency compared to a very significant difference (P<0.01); two acupuncture treated group of rats before and after the treatment in the mean escape latency has significant difference (P<0.01);(2) Mean across frequency Between the groups:treated with EAON, two groups of rats on average across the platform frequency compared with model group increased obviously, the difference was significant (P<0.01); Within the groups:model control group, two rats in group treated with electro made this group mold and on average across the platform comparisons, with significant difference (P<0.01); acupuncture therapy for4weeks rats before and after the treatment in the average number of crossing platform had very significant difference (P<0.01);2.Morphological changes of tissues:4weeks treatment group compared with2weeks group and the model group improved significantly, hippocampal neurons are more regular, nuclear volume increase, prominent nucleoli, inflammatory cell number decreased, while the neuronal pyknosis and hyperchromatic phenomenon of lighter;3.Changes of neuronal ultrastructure:4weeks treatment group compared with2weeks group and the model group improved significantly, neuronal cells can form, Nissl bodies are relatively small, visible lipofuscin, free ribosomes number distribution is normal, synaptic boundary is clear, the number of synaptic vesicles, partial mitochondrial vacuolar degeneration;4.The positive expression of Cholinergic neurons:(1) ChAT model control group, the positive expression of CHAT decreased significantly, and significant differences compared with the blank group and the sham lesion group (P<0.01);CHAT-positive of the4weeks EAON group was significantly enhanced,compare with the2weeks EAON group and model control group,there are obvious differences (P<0.01);(2) ChATmRNA model control group,the positive expression of ChATmRNA decreased significantly, and significant differences compared with the blank group and the sham lesion group (P<0.01);ChATmRNA-positive of the4weeks EAON group was significantly enhanced,compare with the2weeks EAON group and model control group,there are obvious differences (P <0.01);(3) activities of AchE model control group, AchE positive expression was significantly lower, and the blank groupand sham group differences are significant (P<0.01);AchE-positive of the4weeks EAON group was significantly enhanced,compare with the2weeks EAON group and model control group,there are obvious differences (P<0.01);5.The positive expression of monoamine neurotransmitter:(1)5-HT model control group, the positive expression of5-HT decreased significantly, and significant differences compared with the blank group and the sham lesion group (P<0.01);5-HT-positive of the4weeks EAON group was significantly enhanced,compare with the2weeks EAON group and model control group,there are obvious differences (P<0.01);(2) DRD1model control group,the positive expression of DRD1decreased significantly, and significant differences compared with the blank group and the sham lesion group (P<0.01);DRD1-positive of the4weeks EAON group was significantly enhanced,compare with the2weeks EAON group and model control group,there are obvious differences (P<0.01).Conclusion:1.EAON can shorten the AD rats and the mean escape latency, increased the average times of striding over platform, improving compound AD in a rat model of learning and memory.2.EAON can improve the cell structure, reducing the number of inflammatory cell, so that the remaining neurons play a greater role, that acupuncture can protect nerve cells in the brain.3.EAON can make the necrosis of the marginal zone of the increase in number of synapses, and enhance nerve conduction, composite AD model rat learning and memory impairment to improve the function of.4. EAON can improve the composite AD model rat hippocampal tissue of cholinergic neurons of ChAT in the expression,and make the positive expression of the ACHE weakened to facilitate the restoration and regeneration of damaged nerve.5.EAON can enhance the composite AD model rat hippocampus monoamine neurotransmitter expression(5-HT,DA), increased monoamine neurotransmitter secretion, thereby preventing the AD in a rat model of ability of learning and memory. 6.EAON treatment for4weeks group therapy is more effective than EAON treatment group in2weeks.
