Skeletal muscle disuse atrophy (SMDA) has been one of the major issues in research erea of clinical medicine and rehabilitation medicine. It is rare for skeletal muscle disuse atrophy of the rehabilitation process with the application of vibration of this new stimulus intervention recovery measures in the domestic and foreign literatures.Objective:This study was based on animal experiments, to compare the effect of the different frequencies of vibration stimulation within safe range on disuse atrophy of skeletal muscle rehabilitation, and preliminary study on the mechanism, provide reference basis for application of vibration training in disuse muscle atrophy rehabilitation.Methods:50female SD rats randomly divided into five groups:the control group (A), low frequency vibration group (B), high frequency vibration group (C), suspension group (D), natural recovery group (E). After14days suspension, the rats soleus muscle tissue of group D was taken to be specimens, the other two groups were given vibration stimulation training repectively with low frequency as20Hz and high frequency as30Hz. The control group (A) and natural recovery group (E) maintained on a normal diet, activity. Four weeks after taking4groups of rats soleus muscle tissue for specimens were measured in each group of the wet weight to body weight ratio; And made quantitative analysis of rat soleus muscle type Ⅰ, type Ⅱ muscle fiber ratio, average cross-sectional area (CSA); Using Western blotting method on several muscle protein growth factor as IGF-1, MyoD, Myogenin, Myostatin, TGF-β, bFGF for quantitative analysis and comparison. Explore the role of various muscle growth factors in vibration training for SMDA.Conclusion:(1)Both the low frequency and high frequency of the vibration stimulation can be the effective promotion of disuse muscle atrophy of rat soleus muscle morphology and structure of recovery:wet weight of muscle increases and muscle fibers of type Ⅰ and Ⅱ in the muscle cross sectional area increases, can effective against transformation in type of muscle from type Ⅰ to type Ⅱ.(2) Both the low frequency and high frequency of the vibration stimulation can cause the over expression of IGF-Ⅰ, MyoD of the rats in model of disuse muscle atrophy of soleus muscles, and Reduces the expression of Myostatin.It can be the promotion of inhibition on skeletal muscle atrophy by increasing skeletal muscle protein synthesis and decreasing skeletal muscle protein breakdown.(3) The effect of rehabilitation by vibration stimulation in different frequencies has certain difference, integrated morphological and skeletal muscle protein expression results of determination to carry on the analysis, in which the20Hz low-frequency vibration stimulation effect is more obviously.(4) The muscle growth regulator under different frequency vibrations stimulate like MyoD and Myogenin, Myostatin and, TGF-β was not consistency, as they were belong to the same family,Research proposal:(1) As an effective rehabilitation intervention, appropriate frequency of vibration stimulation can be used for one of the rehabilitation therapy of disuse muscle atrophy.(2) For prognosis of and the observation of the,intervention in disuse atrophy of skeletal muscle. According to the results, IGF-1, MyoD and Myostatin can be used as ideal molecular biology application...
