Object: To establish models of stress-restrain ulcer with cold and hot syndrome. Reveal the regulation of stress-restrain ulcer models with cold and hot decoction (Dahuang Huanglian Xiexin Decoction and Lizhong Pill). Provide evidence for further research on stress-restrain ulcer with cold and hot syndrome.Methods: Use cold factor(cold water,0.3mol/L NaOH) and hot factor(8%pepper powder,60% ethanol mixed solution) to establish the model of cold and hot syndrome combined with water immersion stress method which has been received by domestic and abroad scholar, and treat with Dahuang Huanglian Xiexin Decoction of cold property and Lizhong Pill of hot property. 1. Killed the rats seven hours after water-stress. Detected the ulcer index, the content of VEGF, TXB2,PAF,NT,COR and cAMP in blood serum. 2. Detected the EGF, TGF-а, SOD and MDA in the stomach tissue. 3. Detected the content of 5-HT and NE in the brain tissue. 4.Use Immuno-histochemistry to detect the express of Bax and Bcl-2.Results: 1.Compared to the blank group, the content of COR,TXB2,bFGF of the cold syndrome with water-stress group and the hot syndrome with water-stress group increased( P<0.01or P<0.05) .The content of TXB2 in the cold syndrome with water-stress group treated with Lizhong Pill and the hot syndrome with water-stress group treated with Dahuang Huanglian Xiexin Decoction were decreased apparently compared to the cold syndrome with water-stress group and the hot syndrome with water-stress group respectively(P<0.01)compared with Dahuang group, the COR increased(P<0.01),while the content of bFGF decreased (P<0.05) .2.Compared to the blank group, the content of AchE in the cold syndrome with water-stress group and the hot syndrome with water-stress group decreased ( P<0.01 or P<0.05).The content of AchE in hot syndrome with water-stress group was lower than that of the cold syndrome with water-stress group. The content of AchE in the cold syndrome with water-stress group treated with Lizhong Pill and the hot syndrome with water-stress group treated with Dahuang Huanglian Xiexin Decoction were increased apparently compared to the cold syndrome with water-stress group and the hot syndrome with water-stress group respectively(P<0.01 or P<0.05).3.Compared to the blank group, the content of cAMP in the cold syndrome with water-stress group was decreased ( P<0.01 or P<0.05).Compared to the blank group,the content of cAMP in the hot syndrome with water-stress group was increased(P<0.01 or P<0.05) .Compared to the cold syndrome with water-stress group, the content of cAMP was increased in the cold syndrome with water-stress group treated with Lizhong Pill. Compared to the hot syndrome with water-stress group, the content of cAMP was decreased in the hot syndrome with water-stress group treated with Dahuang Huanglian Xiexin Decoction. 4.Compared to the blank group and hot syndrome with water-stress group, the content of PGE2 and NT were decreased in the cold syndrome with water-stress group ( P<0.01 or P<0.05) .Compared separately to the cold syndrome with water-stress group treated with Lizhong Pill and the hot syndrome with water-stress group treated with Dahuang Huanglian Xiexin Decoction, the content of PGE2 and NT were increased in the cold syndrome with water-stress group and the hot syndrome with water-stress group ( P<0.01).5.Compared to the blank group and cold syndrome with water-stress group, the content of VEGF and PAF were increased in hot syndrome with water-stress group ( P<0.01 or P<0.05) .Compared to hot syndrome with water-stress group, the content of VEGF was increased and the content of PAF was decreased in the hot syndrome with water-stress group treated with Dahuang Huanglian Xiexin Decoction( P<0.01 or P<0.05).6.Compared to the blank group, the content of Bcl-2 was decreased in the cold syndrome with water-stress group and the hot syndrome with water-stress group,while,the content of Bax was increased. Compared with cold syndrome with water-stress group and the hot syndrome with water-stress group separately, The content of Bcl-2.in the cold syndrome with water-stress group treated with Lizhong Pill and the hot syndrome with water-stress group treated with Dahuang Huanglian Xiexin Decoction were increased apparently, while the express of Bax was decreased. Conclusions: 1.Hot syndrome with water stress ulcer model has relationship with the increasing of bFGF,TXB2,COR,VEGF,PAF,cAMP,and has relationship with the decreasing of AchE. And it also has relationship with the decreasing of Bcl-2 and the increasing of Bax.2.Cold syndrome with water stress ulcer model has relationship with the increasing of bFGF,TXB2,and has relationship with the decreasing of AchE,PGE2,NT,cAMP.And it also has relationship with the decreasing of Bcl-2 and the increasing of Bax.3.The mechanism of the cold formulae, Dahuang Huanglian Xiexin Decoction, to prevent the water-stress induced ulcer with hot syndrome has relationship with increasing of VEGF,decreasing the content of PAF,stimulating the releasing of bFGF,inhibiting AchE,reducing TXB2,reducing the COR by inhibiting the activity of sympathetic adrenal.4.The mechanism of the hot formulae,Lizhong Pill,to prevent the water-stress induced ulcer with cold syndrome has relationship with increasing of cAMP,decreasing the content of TXB2,stimulating the releasing of bFGF,decreasing the activity of AchE, improving the content of PGE2 and NT,decreasing the death of cells when in stress situation.
