| This is a new mutation source which named high-energy particle field was selected to irradiate Longmu 803 alfalfa dry seeds with 5 different dose(109Gy,145Gy,195Gy, 284Gy,560Gy), with the same dose,60Co-γray was selected to irradiation another 5 group Longmu 803 alfalfa dry seeds. The data of plant quality, plant weight and heritability between generations were collected to compare with each other, we need compare the plant data in same mutation sources but with different dosages and same irradiation dosage but in different mutation sources group. We also compared the Ability of the second future generations of cold tolerance. Then we using Diode Array 7200 near-infrared reflectance spectroscopy (NIRS) established calibrations of crude protein and crude fiber in 152 alfalfa samples, which were developed by means of partial least squares (PLS) regression on the basis of chemical analytical data. We set lnm,2nm and 5nm interval wave separately and two kinds of samples (crude and fine), accuracy and repeatability were verified so as to compare superiority and inferiority. The result shows:1 In M2, The heritability of the CR group is better then the 60Coγ-rays group at the same dose; in the same mutation sources group, higher dosage has better heritability than lower dosage, the result of heritability data also shows CR284 is better than all other dosage, has the better effect for choose the early generation2 Only the plant without frostbite be chosen to test the Ability of the second future generations of cold tolerance. At last only 3 dosages can be collected leaves for the test. We test the effect of M2 on Longmu No.803 alfalfa subjected to low temperature treatment between autumn and winterwere studied, including monitoring the leaf for free proline content, SOD activity, LSSC, Chla and Chlb contents after treatment with different radiant dosages. The mixed high-energy particle field significantly (P<0.05) increased the cold tolerance of M2 compared with the control. With increasing radiant dosage, the proline content, SOD activity, LSSC, Chla and Chlb contents of M2 were all significantly reduced. The lowradiant dosage induced stronger cold tolerance than the high radiant dosage. There were two options involving145 or 195 Gy of mixed high-energy particle field intensity.3 About the NIRS, the main conclusions of this part includes: 1) Through spectrum scanning to 152 different samples at 3 different interval wavelength, Results show that the best validation result is in 950-1650nm wavelength;2) Because protein is measured by complete-automatic instrument, which metrical precision is higher, so the correlation coefficients of calibration (R2) is higher than fiber and the exceptional point elimination is smaller than fiber;3)Through investigating how smoothness and differential coefficient pretreatment effect on modeling outcome, result shows that model R2 and SECV change little with spectrum smoothness, fine samples obtain most R2 and least SECV in 5,7 and 9 point smoothness, and that crude samples spectrum smooth disposal as well as 1stDeriv and 2ndDeriv differential coefficient founded modal effect are all not as good as not disposed samples;4)Through investigating how fine or crude samples and interval wavelength effect on modeling, result show that fine samples in 2nm obtain the most R2,F value and the least SECV,SEP, difference in SECV and SEP is also the least;Basing above analysis establishment, the correlation coefficients of calibration (Rcal2) were 0.97,0.94 for crude protein, crude fiber, the correlation coefficients of validation(Rval2) were 0.96,0.92, the SECV of these parameters were 0.42,0.78, it was better than lnm and 5nm. The experiment showed that there were no obvious differences in NIRS and standard methods; The fine samples'accuracy is above crude samples; At the same time, it provided a new model to verify the compositions of alfalfa' crude protein and fiber. In conclusion, it is feasible that NIRS analysis method apply on measure alfalfa's crude protein and crude fiber content. At the same time, modeling units can be enlarged unceasingly by new samples and be made complementary and perfect with added data.Otherwise, I carry on field investigation to this 152 individual strains'2007 year return green strains and registered phenotype's data, using RAPD molecular maker to investigate DNA polymorphism to obtain molecular data. So it can be become evidence of seed selection.From the analysis of variance(ANOVA) of which different disposals effect on different varieties'fiber and protein content, I conclude different disposals have notable effect on fiber content of zhaodong alfalfa, longmu801 alfalfa, but there is little effect on longmu803 alfalfa; Different disposals have notable effect on protein of longmu801 and longmu803, have little effect on zhaodong alfalfa. It coincides with phenotype analysis above on the whole. Different disposals have little effect on else phenotype character(escape plant height), it has some effect on plant height.From the analysis of variance of molecular and phenotype data, I conclude that OPA9-6, OPD10-5, OPX13-6 three leading sequence's coordinative location have notable difference from the content of protein. There is no linkage location from the content of fiber.From the analysis of correlation of every phenotype data, each phenotype character has high correlation except very few character, for example there is no relativity fiber and plant weight, the second fresh weight. This can be taken as evidence of each character linkage tightness degree and provide evidence for seed selection.From the analysis of relativity of molecular and phenotype data, I conclude that molecular data and fiber, plant height, dry height have notable relativity, molecular data and protein content have comparative notable relativity.From clustering analysis of molecular and phenotype character, I conclude that longmu801 and longmu803 cluster two sorts separately, and that zhaodong alfalfa genetic polymorphism rule is close to longmu803. The same sort's different disposal dose cluster together separately, it illuminate that there is high relativity between alfalfa species genetic polymorphism and disposal doses. In longmu801, the variance range change little along with disposal dose. Because variance varieties are fewness, and be clustered to other sorts or became one sort by itself. |
