1. Study on the inhibition effect mechanism of Sorbaria sorbifolia chloroform extract in S180 mice. The S180 sarcomas was transplanted in 60 KM mice, then they were divided randomly into 5 groups to receive different treatment (high, medium and low dose group of Sorbaria sorbifolia chloroform extract, cyclophosphosphamide group and control group). Then 10 days later, execute the mice. The inhibition ratios of the tumor, the spleen index and thymus index were calculated out, the activities of NK cell and the contents of TNF-α, IL-2 in serum were detected. And the levels of VEGF and Ang-1 proteins were assayed with RT-PCR and Western blot. The results showed: the Sorbaria sorbifolia chloroform extract could increase the levels of the inhibition ratios of the tumor weight and the spleen index and thymus index in mice. The activities of NK cell and the contents of TNF-α, IL-2 in serum were significantly higher than that of control group. It also could markly downregulate the expression of VEGF gene, compared with control group. Yet the expression or Ang-1 protein has no remarkable differences in each group.2. Inhibition of 5, 2', 4'-trihydroxy-6, 7, 5'-trimethoxyflavone on proliferarion of hepatoma HepG-2 cell line. 5, 2', 4'-trihydroxy-6, 7, 5'-trimethoxyflavone inhibited the proliferation of HepG-2 cell and the effects were in a time-and-concentration dependent manner by MTT assay. 5, 2', 4'-trihydroxy-6, 7, 5'-trimethoxyflavone can also induced apoptosis, the G2/M phase was arrested and the ratio of apoptosis was significantly different as compared with that of control group, respectively. The expressions of apoptosis-associated gene bcl-2, bax and p53 had changed significantly, respectively.3. Studies on chemical constituents in Sorbaria sorbifolia. The constituents from Sorbaria sorbifolia chloroform and ethal acetate extracts were isolated with sillia gel column chromatography and identified by their physical, chemical constants, spectral analysis and comparison with the authorized samples. Severn compounds were isolated and identified as 5, 2', 4'-trihydroxy-6, 7, 5'-trimethoxyflavone (Ⅰ), 5, 7-dihydroxy-8-methoxyflavone(Ⅱ), rutin(Ⅲ), Quercetin(Ⅳ), daucosterol(Ⅴ), benzoate (Ⅵ) and p-hydroXybenzoic acid (Ⅶ). Severn compounds were isolated from Sorbaria sorbifolia chloroform and ethal acetate extracts for the first time.
