| Aim: To investigate the effects of 17β-estradiol (E2), progesterone (P4) and human chorionic gonadotrophin (HCG) corresponding to pregnant concentration on infection and replication of HTV-1 in vitro.Methods: To research effects of HCG in final concentration of lOU/mL and 100U/mL, and E2 or P4 in final concentration of 10-7mol/L, 10-6mol/L and 10-5mol/L on HIV-1 infection and replication in vitro by HIV-1 mediated fusion and ELISA detected concentration of HIV-1 p24 antigen respectively. Besides, effects of E2, P4 and HCG corresponding to pregnant concentration on lymphocytic activation and proliferation were determined by detecting CD69 and CD25 expression by multicolor flow cytometric analysis and the 3H-thymidine (3H-TdR) incorporation respectively.Results: P4 at concentration of 10-6mol/L and 10-5mol/L, and E2 at placental regional level of 10-5mol/L, as well as HCG of 100U/mL could dramatically inhibited formation of fusion mediated by HIV-1. The suppressing effects of E2 and P4 were identical with their dosage. HCG of 100U/mL and E2 or P4 at placental regional concentration of 10-5mol/L could decrease releasing of p24 antigen. E2 and P4 at pregnant level of 10-6mol/L and 10-5mol/L, as well as HCG of 100U/mL inhibited early activation of T lymphocyte and CD4+T cells, and suppressing effects of E2 and P4 were in dose depended. E2 or P4 of 10-5mol/L inhibited expression of CD25 as a late activated molecular. E2 and P4 at concentration corresponding to pregnant level, and HCG of 100U/mL decreased incorporation of 3H-TdR of PBMC stimulated by PHA, and depressive effect of P4 on proliferation was in dose depended.Conclusion: HCG, E2 and P4 in concentration detected at the fetal-maternal interface could inhibit not only infection and replication of HTV-1, but activation and proliferation of lymphocytes in vitro.
