| Obesity, defined as excess adipocytes number and cell size, results from imbalance between energy intake and energy expenditure. Obesity is a significant risk factor for many serious illness, resulting in reducing people health. Based on the study that the pomegranate leaf tannides (PLT) had a noticeable activity of modulating dyslipidemia of hyperlipidemia rats, this study was examined the effect of PLT and its main components SY1, SY2 and SY3 on the adipocytes lipid accumulation and preadipocytes differentiation based on three kind of levels which were cell level, molecule level and gene level and finally found the principal bioactive components of PLT. The PLT, SY1 and SY2 decreased the adipocytes uptaking the TG from the plasma by inhibiting the LPL activity and decreased the intracellular lipolytic activity by inhibiting the fatty acid and glycerol released out intracellular. The bioactive component of the suppressing effect in adipocytes lipid accumulation was SY1. This study was also found that the PLT and its bioactive component SY2 had inhibited the preadipocyte differentiation, inhibited the glycerol-3-phosphate dehydrogenase (GPDH) activity, the key enzyme involved in the cell differentiation and suppressed the PPARγ mRNA transcription and expression. The intracellular pharmacokinetic study showed that the two componentsSY1 and SY2 could transport into adipocytes. Finally, three different models were confirmed that the PLT could suppressed the increase in fat tissue weights in the high-fat diet induced obese mice. It was found that the PLT strongly inhibited the lipid absorption from the intestinal to reduce blood triglyceride levels, which might be another mechanisms of anitobesity effects by PLT. |
