Objective Acute myeloid leukemia (AML) is an aggressivemalignancy that generally leads to a poor outcome.The complete remission(CR) after intensive chemotherapy and allogeneic stem cell transplantation(allo-HSCT) is high,but the prognosis still poor. The further investigation for new treatment strategies and drugs will require detaild knowledge about the regulation of leukemic hematopoiesis by the cytokine in the bone marrow.Leptin is a regulator of fat metabolism that is synthesized in adipocytes,and was shown to stimulate myeloid proliferation.The leptin receptor(OB-R) was highly expressed in AML,maybe act a pole in pathophysiology of AML.Methods By RT-PCR, the expression of PYST2 mRNA of 43patients with newly diagnostic AML, 17 with ALL,6 with relapsed AML, 13 in complete remission, 8 healthy controls and 5 cord blood mononuclear cells was assayed. The relationships between expression of OB-R mRNA and patients' clinical characteristics were evaluated.Results Both the long (OB-R-_L)and short (OB-R-s) isoforms ofOB-R was low expressed in normal BM and cord blood; OB-R-_L and OB-R-s that was detected is 69.76% and 51.16% with newly diagnostic AML,which was higher than ALL(P=0, 0.006);both were expressed in...
