The research, work on cancer gene therapy has made a great progress since 1989. Using cytokine transfected tumor cell as vaccine toinduce anti-tumor immunity in vivo for the inhibition of certain malignant tumors growth was a hot point in current cancer immuno- or gene therapy. For improving it's therapeutic efficiency, one of the important measures may be considered of administrating the novel comprehensive treatment strategy. In this study, IL2 was introduced into B16 cell line established originally from a melanoma in mice, and bearing a highly malignant and metastatic but poorly immunogenic behavior. The effect of B16 cells transfected with IL-2 gene( B16-IL2) on the immune reaction of mice was studied systematically in order todetect the mechanism of B16-IL2 cells in promoting the immunity ability against the development of malignancies as well as to study the effectof B16-IL2 cells on the induction of cytotoxic T lymphocytes(CTL) in vitro, which is known to be specific against B16 melanoma. It is expected to get a better and more efficient result by the combined administration of B16-IL2 vaccine and CTL at the same time rather than by using B 16 - IL2 vaccine alone for the immunotherapy of tumor-bearing mice.Retrovirus infection was adopted for the transfection of... |
