Experimental Research Of Effect Of Shenshuaibaoshen Capsule On Mouse Renal Fibroblasts

Objective: To elucidate the mechanism of Shenshuaibaoshen Capsule delaying evolve of chronic renal failure and interstitial renal fibrosis by observing the effects of Shenshuaibaoshen Capsule on proliferation and the variations of LN and Col IV and cytokine mRNA expression of mouse renal interstitial fibroblasts, and to provide the experimental evidence for Professor Zhangqi ' s traditional Chinese medicine theory of FUFANGDUOBADIAN. Methods: The cells were cultured in media containing various contents of SBS Capsule.The Proliferation was assayed with MTT method.The variations of LN and Col â…£ were assayed with radioimmunoassay method, and TGF-β₁ was assayed with ELISA method.we detected a—SMA, Vimentin and TGF-β₁ gene expression with immuno -histochemistry techniques.We observed the expression of ICAM — 1mRNA and TGF-β₁mRNA throught semi-quantitative RT-PCR. Results: The level of LN, Col â…£ and TGF-β₁ in the cell cultivation liquid of the SBS team is lower than in the cell cultivation liquid of the contrast team. SBS could relieve immuno-histochemical expression of Col â…£ and Vimentin and TGF-pi in the renal fibroblasts.SBS had a restraining effect on proliferation with dose-dependence and time-dependence , and induced an decreasing expression of ICAM—1 mRNA and TGF-β₁ mRNA with dose-dependence to the renal fibroblasts. Conclusions: Shenshuaibaoshen Capsule can delay chronic renal failure and interstitial renal fibrosis by restraining effect on proliferation to the renal fibroblasts, and inhibiting the ICAM— 1mRNA, TGF- β₁mRNA expressions in the renal fibroblasts and reducing Colâ…£, LN.
