Objective: To discuss the mechanism of ventricular remodeling in viral myocarditis caused by repetitive infection and the effect of MMP family on ventricular remodeling and the relationship between the MMP family and the correlative cytokines. To research the protection and the mechanisms of the traditional Chinese medicine to the myocardium injury of myocarditis. Method: To build the model of viral mycocarditis which is formed by mice those were infected repetitively 4 times by CVB3m intraperitoneally with a gradient double dose. The treatment team was given medicine right after the first inoculation and last to 60 days.To observe the change of myocardium structure, heart quality index, degeneration and necrosis of myocardium collagen hyperplasia in myocardium interstitial respectively via microscope at different time after the final infection. Also, RT-PCR was used to determine mRNA level of MMP-1, TIMP-1, TNF-α, TGF-β1 in myocardium. Western blot were used to measure protein content of MMP-1, TIMP-1 in myocardium. ELISA was adopted to measure TNF-α, TGF-β1 level in serum.Result: Pathology analysis shows that the myocardium degeneration of traditional Chinese medicine team and Western medicine team were mitigate obviously to the virus infection comparison team and also dose the heart quality index. At viral myocarditis acute period (5 to 10 days after final infection), myocardium necrosis and inflammation cells soakage were relatively much, the transcription and translation level of TNF-α and MMP-1 expression were obviously increased. The treatment teams inhibit the forepart expression of TNF-α and MMP-1. At chronic period (since 20 days after final infection), myocardium necrosis focus has been absorbed gradually and collagen hyperplasia evidently, the expression of TGF-β1 in serum and the level of TIMP-1 in myocardium increased. The analyses ofthe treatment teams show that the medicine can depress the level of and TIMP-1. They all have statistics difference contrast with the virus infection comparison team.Conclusion: Successfully built the model of myocardium necrosis and ventricular remodeling in viral myocarditis caused by repetitive infection, approved that the method has positivity effect to CVB3-VMC and increase the livability of VMC mice. The method can inhibit the myocardium collagen hyperplasia, depress the level of gene and protein, improve ventricular remodeling, regulate the balance of MMPs/TIMPs via inhibit TNF-cc and TGF-pi and suspend myocardium fibrosis.Originality: Pathological changes and collagen hyperplasia of myocardium and how MMP family changed and was regulated in myocarditis caused by repetitive CVB.i,,, infection were observed for the first time. And first research The protection of the traditional Chinese medicine to the mice myocardium injury and myocardium fibrosis. Applying molecular biology method to observe the effect about traditional Chinese medicine to the cytokine, MMPs system and the course of ventricular remodeling.
