| For the past few years, along with the increasingly deep investigation of Peroxisome proliferator-activated receptor gamma 2 (PPARγ2)—which is the key transcription factor of adipocyte differentiation,we found it may be playing a important role in bone metabolism process.As marrow adipocyte and osteoblast are derived from a common multipotential mesenchymal stem cells (MSCs) progenitor,and there quantum in bone marrow have an inverse relationship,cohere with the evidence that various kinds of osteoporosis all have increasing adipocyte in bone marrow,we shoud pay more attention to the relationship between the activation of PPARγ2 of bone marrow cells and bone metabolism .Mounting evidence indicats that during aging,the potency of differentiation to adipocyte of MSCs is increased, the modulator of this process PPARγ2 has interaction with the transcription factor of osteogenesis—Core binding factorαl (Cbfαl), high expression of PPARγ2 can reduce cytopoiesis of osteoblast in bone marrow which may effect skeleton formation, This may be a reason of osteoporosis and fatty marrow .The osteoclast (OC) which mediate bone resorption and play a key role in age-related osteoporosis is derived from hematopoietic stem cells (HSCs) ,and it is HSCs but no MSCs express PPARγ2,the relationship between the activation of PPARγ2 of bone marrow cells and osteoclast is less well studied and rather controversial, so it worth further research.The reason of increasing adipocyte in bone marrow has not been identified,whether or not it related to the augmented endogenic ligand of PPARγ2 with aging is not yet know; otherwise, someone are worry about the clinical application of thiazolidinediones–synthetic agonist of PPARγ2 which are indicated for use in type 2 diabetes mellitus could induce negative effect in bone metabolism, therefore, we planned this essay to identify the possible agent of high expression of PPARγ2 in bone marrow with aging and to identify the relation between PPARγ2 and differentiation and activation of osteoclast, expected to offer a new sight in treatment of osteoporosis and to give a theoretical guidance to TZDs in clinical medication.Partâ… :The relationship between activation of PPARγ2 by endogenic ligand in bone marrow and typeâ…¡primary osteoporosis in aging rats.Objective: Through observing the inter-relation of PPARγ2,Ox-LDL and bone loss in... |
