Aseculfame K is an antificial sweetener which has been used in the food industry for some years. As yet no metabolic effects have been found. It. was reported that the sweetener can induce a cephalic (?)hase of insulin secretion. To ana(?)yse the mechan(?)sm of this (?)enomenon. the effect of Acesulfame K on insul in secretion in vivo and in vitro had been stedied.IN VIVO EXPERIMENTS:1. Cephalic ohase: Usino the undistrubled and free-movino rats. the effect of Acesulfame K on cephalic (?)hase insul in secretion was studied. After the rat drank 28% (?)lucose or0.2% Acesulfame K. (?)asma insulin increased sionificant1y in the first minute, without any chanoe in blood (?)lucose at the same time. D(?)inkino 0.5% Saccharin. 0.9% Nacl and tap water had no sianificant influence neither in plasma insulin concentration nor in blood olucose level.2. Injection and infusion: Male Wistar rats, weighing 250-300-were fasted overni(?)ht and anaesthetized_withpentobarbital. A silicon catheter was inserted into theri(?)ht cervical vein for inhection of glucose. Acesulfame Kand olibenclamide , and for obtaininc. blood samples. Insome experiments, another catheter was inserted into theleft cervial vein for continuous infusion. Blood samples weredrawn at 0. 5. 10. 15. 30 and 68 min after injection.and at -10. 0. 10, 20, 30, 40, 60, 80, 100 and 120 min whenthe infusion started.
