Objective: To study the expression of Rho-kinase (ROCKI and ROCKII) in the development of hypoxic pulmonary hypertension and the effect of Breviscapine on Rho-kinase. To investigate the role of Rho-kinase in hypoxic pulmonary hypertension and the mechanism of Breviscapine on hypoxic pulmonary hypertension. Methods: Forty two adult male SD rats were randomly divided into seven groups. One group was exposed to air (normal group) and the other five groups were exposed to isobaric hypoxia for 1 day, 3 days, 1 week, 2 weeks and 3 weeks respectively. And the last group was exposed to hypoxia for 3 weeks and treated with Breviscapine at the same time. A cardiac catheterization was used to measure the mean pulmonary arterial pressure (mPAP). Separating and weighing the right ventricle (RV) and left ventricle plus atrial ventricular septum (LV+S) to calculate RV/(LV+S). The ratio of thickness/vascular external diameter (WT%) and the ratio of area/total vascular area (WA%) were measured by image analysis. The quantity of Rho-kinase and Rho-kinase mRNA in rat pulmonary arterioles were determined by immunohistochemistry, in situ hybridization and image...
