Objretive: 1. To observe the effects of Rouganyixianyin Decoction in preventing the form of liver fibrosis caused by infection of schistosoma japonicum, and compared the effects with Colchicine which was took as the control-drug. 2. Try to find the difference of the effects in preventing the form of liver fibrosis between Rouganyixianyin Decoction only and the Decoction combined with Erzhi pill, thus reveal the mechanism of anti-fibrosis.Methods: Zoopery 1: The earlier stage of mice hepatic fibrosis model was made by infected with cercariae of schistosoma japonicum, 60 mice were divided into 5 groups: group A (Normal control group), group B (Model group), group C (Praziquantel group), group D (Rouganyixianyin group), group G (Colchicine group), mice of each group were infected with cercariae except group A. From the 21st days after infection, group D were fed with Rouganyixianyin Decoction, group G with Colchicine solution, group A,B,C were fed with physiological saline respectively for 40th days. Praziquantel solution were fed to the mice of Group C,D,G merely on the 45th days of infection. Mice of each group were put into death on the 61st days, tested the indexes of hepatic function(ALT & ALB) , the contents of HA,PCⅢin serum, HYp in liver tissue. The histopathology of hepatic tissue was observed too (by HE/VG dye, electron microscope, and immunohistochemical methods), The expression of collagen typeⅠand typeⅢwere measured by quantitative analysis in order to reflect the composition of ECM.Zoopery 2: The earlier stage of mice hepatic fibrosis model was made by infectedwith cercariae of schistosoma japonicum, 48 mice were divided into 4 groups: group A (Normal control group), group C (Praziquantel group), group D (Rouganyixianyin group), group E (Rouganyixianyin + Erzhi-pill group), mice of each group were infected with cercariae except group A. In terms of lower ALT and HA, increase ALB, improve indexes of liver & spleen, quantitative analysis of VG dye and collagen typeⅠ, there were no significant difference between Rouganyixianyin group and Colchicine group(P>0.05); but for PCⅢ,Hyp and the quantitative analysis of collagen typeⅢ, Rouganyixianyin was much better than Colchicine(P<0.05). It shows that Rouganyixianyin Decoction has confident effect on preventing the form of liver fibrosis caused by infection of schistosoma japonicum, the Decoction can be used as the instead drug of Colchicine in future. 2 Group D (Rouganyixianyin group) and group E (Rouganyixianyin + Erzhi-pill group) were all had much better effect than Group C (Praziquantel group) to improve indexes of spleen, decrease ALT,HA level in serum, resist the expression of collagen typeⅠand typeⅢ(P<0.05, P<0.01), But there were no significant difference between Group D (Rouganyixianyin group) and group E (Rouganyixianyin + Erzhi-pill group). Observations of liver histopathology also refers that Rouganyixianyin only or combined with Erzhi-pill had remarkbale effects on anti-liver fibrosis. However, for improve indexes of liver, increase ALB, decrease HYp, resist the expression of TGF-β1,a-SMA, Rouganyixianyin combined with Erzhi pill was much better than Rouganyixianyin Decoction only(P<0.05).Conclusions: According to the theories of TCM, we holds that the pathogenesis of liver fibrisis were due to deficiency in origin and excess in superficiality, deficiency in origin mainly refers to deficiency of liver-yin and kidney–yin, blood stasis obstructed inliver collaterals was the main fact of excess in superficiality. Deficiency of qi and blood in the liver collaterals resulted in protracted course of disease. Nourishing yin and promoting blood flow, thus activating liver collaterals are the important treatment of liver fibrosis.Rouganyixianyin Decoction has confident effect on preventing the form of liver fibrosis caused by infection of schistosoma japonicum, the Decoction can be used as the instead drug of Colchicine in future. Rouganyixianyin combined with Erzhi-pill also had remarkbale effects on anti-liver fibrosis, it can improve indexes of liver, increase ALB, decrease HYp, resist the expression of TGF-β1,a-SMA, etc. The effect of Rouganyixianyin combined with Erzhi pill was proved much better than Rouganyixianyin Decoction. The effects of the two Decoctions can be concluded as: resisting the expression of TGF-β1 ; inhibiting hepatic satellite cells activation; decreaseing the production of type I and type III collagen; promoting the degradation of collagen; preventing the form of hepatic sinusoid capillarization. The anti-fibrosis mechanism of Erzhi pill and it's compositions is not very clear yet, needs more research in future.
