| Oxyphenamone (Oxy) is a new cardiovascular active compound synthesized by Institute of Materia Medica. It has both positive cardiotonic and vasorelaxations effects. It acts as an inotropic agent without raising intracellular Ca2+ levels and belongs to a kind of calcium sensitizers. However, the mechanism of its vasorelaxations is not very clear. For further clarifying the vasorelaxations of Oxy, we tested its effects on isolated small arteries preparation of rabbit, and studied the molecular mechanism of its vasorelaxation with vascular smooth muscle cells which primary cultured or isolated from small mesenteric arteries of rabbit.METHODS1 Bioassay: Helical strips prepared from rabbit renal artery, femoral artery and mesentery artery were studied in isometric myographs.2 Confocal laser scanning microscopy (Argon Ion): Calcium signaling was observed in primary culture of rabbit vascular smooth muscle cells dyeing with Fluo3·AM, recording fluorescent changes after the stimulation with Oxy in real time.3 Whole-cell patch clamping: Whole cell currents were recorded in freshly isolated rabbit mesentery artery smooth muscle cells via the whole-cell patch clamp technique. |
