Objective: To explore the use of specific small interfering RNA(siRNA) to suppress Smad (Smad2, Smad3) gene expressions in human keloid fibroblasts, and investigate the effect of Smad siRNA on types I and III procollagen expression. Methods: Smad specific siRNA was designed and the keloid fibroblast were transfected. The expression levels of related genes were examined by RT-PCR and Western Blot. Results: Endogenous Smad2 expression were blocked efficiently in keloid fibroblasts by Smad siRNA. mRNA levels of types I and III procollagen were also significantly and uniquely decreased following the reduction of Smad by siRNA in keloid fibroblasts. Conclusion: The results indicate that Smad(Smad2, Smad3)plays an important role in the TGF-β induced fibrosis in keloid. Downregulation of Smad expression in keloid fibroblasts can significiantly dicrease procollagen gene expression.
