The Experimental Study On Curative Effect Of Prostate Water Pellets Ⅰ Against Chronic Bacterial Prostatitis Of Model Rats

Objective: To investigate the effects of Prostate Water Pellets I (PWP I ) against Chronic Bacterial Prostatitis(CBP) and its mechanism.Methods: we have done experimental researches by using modern scientific methods on levels of body , cell and molecule. Totally 18 male Wister rats were randomly devided into 6 groups: the normal group, the model group, the positive controlled group, the high dosage group, the middle dosage group, the low dosage group, of 20 rats in each group.A rat model of CBP was developed by injecting bacillus coli into the prostate of rat. A week later, high,middle and low dosage of PWP I suspension was perfused into their stomachs .The positive controlled group rats taked Minocycline Hydrochloride Capsules and the others was filled normal saline into their stomachs.After taking medicine for 35 days, we counted colony in the prostate homogenate and measured the levels of IL-2 , IgG and TNF-a in serum with two-antibody enzyme-linked immunosorbent assay. HE staining was used to observe the change of prostate tissue. We observed apoptosis with terminal transferase deoxyuridine nick-end assays and tested NO of the prostate tissue with nitrite reduction test then calculated the activity of NOS. The immunohistochemisty test observed the ICAM-1 expression level in the prostate tissue of every groups.Results : PWP I can restraine bacteria in prostate in the rats infected CBP.PWP I can reduc the abnormally high level of TNF-α and raise the level of IL-2, IgG in the rats infected CBP.PWP I can improve the pathology of prostate tissue in therats infected CBP.PWP I can adjust prostate apoptosis in the rats infected CBP.PWP I can reduce the content of NO and the activity of NOS in prostate tissue.PWP I can decrease the expression of ICAM-1 in prostate tissue.Conclusion : The PWP I manifested good effect on CBP .The principal mechanism of PWP I on treating CBP is related with restraining bacteria in prostate;decreasing the levels of proinflammatory cytokine ( TNF- a) in blood;relieving leucocytes infiltration? depressing the hyperplasia of fibrous tissue and adjusted prostate cell apoptosis.
