Neurotrophic factors are naturally-occurring proteins which are essential for neuronal survival and differentiation during development and for the maintenance of normal neuronal function in the adult. The deficiency of endogenous neurotrophic factors is one of the major reasons for the regeneration and function recovery following peripheral nerve injury, and the application of several neurotrophic factors in combination has been becoming an important method to enhance regeneration and function recovery of peripheral nerve. However, three question need further investigation has occurred in the field. The first is the combination of various kinds of exogenous neurotrophic factors and their administration. Due to the selective and crossed effects on neurons, various kinds of neurotrophic factors should be applied in specific combination to produce synergistic biological effects. Furthermore, because the expression level of various neurotrophic factors changed with different amplitude and phase after peripheral nerve injury, so the administration way including applying time and dose of each neurotrophic factor must be determined based on its expressing characteristics to promote nerve regeneration effectively. The second is the detection of expression of neurotrophic factors. As for the previous study of various neurotrophic factors expression, the respective experimental animal, injury model, detection method and quantitation used by individual investigator were scarcely identical, consequently the comparability between the expressions of various neurotrophic factors was greatly influenced. Even for the certain factor, the changing tendency come from different experiment were seldom consistent with each other. So the detection of the co-expression of neurotrophic factors under comparatively uniform condition is especially important for accurate understanding of the dynamic characteristics of neurotrophic factors during peripheral nerve regeneration. The third is the estimate for the structure recovery of regenerated nerve. Current two-dimensional observation is unable to representing the intact anatomical structure of regenerated nerve.NGF(nerve growth factor), CNTF(ciliary neurotrophic factor) and GDNF(glia cell line-derived neurotrophic factor) are three typical neurotrophic factors that have been distinct in the regeneration of peripheral nerve.
