Objective: To explore the possible relationship between the age-related change of p38 and p130 protein expression and the pathological characteristic of senescence-accelerated and brain atrophy in SAMP10 mice and the mechanism of improvement effect on learning and memory deficits of P10 by "YiQiTiaoXue, FuBenPeiYuan " acupuncture method.Methods: Using SAMP10 and normal control SAMR1 as model, applying westernblotting technique to monitoring the age-related protein expression difference of p38 and pl30 in neocortex and hippocampus between R1 and P10. We also observed the acupuncture effect on the protein expression in 8-month old P10 brain.Results:1. In the neocortex and hippocampus of Rl control, p38 and p130 expression exhibited an age-related stably uprising tendency.2. In the neocortex and hippocampus of P10, p38 expression exhibited an age-related fluctuating uprising tendency. On the country, p130 expression exhibited an age-related fluctuating descending tendency. These changes occur at 4 to 6 month old and go on to the following months. This time is very similar to that after a period of normal development times (4 to 6 month), P10 express early onset and irreversible senescence phenotypes. Further more, all the notable changes of the two proteins seem to happen in a coincidence manner and towards opposite direction. The age-related protein expression of p38 and p130 indicate that there may be a possible correlation between them and may directly result in the developing process of senescence-accelerated and brain atrophy which feature SAMP10 mostly and distinguishingly.3. By contrasting the protein level of p38 and p130 in P10 to that of age-matched Rl in the same region of the brain, we found that the variance of the protein level among different age groups of Rl is gentle and not obvious, but sharp and acute among age-matched P10. From 2-month old, the protein level of P10 is dramatically different with that of Rl, and this may contribute a lot to the genesis of the pathological process ofP10.4.
