| In our researches, on the basis of chemical and pharmacological studies, processing principle of the rhizome of Cibotium barometz was discussed. After it was processed, the content of effective constituents increased and the irritant constituents decreased. The best processing technology was studied by orphonogal test to provide the theoretical basis for the researches in quality standard of processed products of the rhizome of C. barometz.Our studies were performed in the following three aspects:1. Chemical researchesInfrare Spectrum of the raw and processed samples of C. barometz were determined. There was no significant difference between the raw and processed samples. But the hair of this plant is significantly different from the rhizome. So the further study on the chemical constituents of this medicinal plant is forwarded.The content of tannins in C. barometz were firstly determined by the casein method and complexometric titration respectively. After it was processed, the content of tannins decreased, as was advantageous to strengthening the effects of promoting blood circulation to remove rheumatism.GC-MS was firstly used to analyze the compositions of the volatile oil in C. barometz and the content of volatile oil decreased after it was processed.The contents of protocatechualdehyde, protocatechuic acid, and 5-hydroxymethyl furfural in C. barometz were firstly determined with HPLC at the same time. After it was processed, the contents of the three compounds increased significantly. And the contents of them in the sample stir-fried with sand were the highest.The irritant constituents in C. barometz were firstly studied with GC-MS. And it was found that the irritant property of the medicinals reduced after processed.2. Pharmacological researchesSince C. barometz has the actions of nourishing the liver and kidney, and removing rheumatism, we designed to study its pharmacological effects in the following aspects: anti-inflammation, analgesia, promoting blood circulation, nourishing the kidney yang, and immunization, etc. It was confirmed to be effective and the processed samples were more effective than the raw one.It was firstly studied to be inhibitory to auricular hematoma in mouse induced by dimethylbenezyl.It was firstly found that C. barometz and its various processed samples were significantly inhibited the platelet aggregation induced by thrombin inrabbits. And the processed samples were stronger in the inhibition than the raw one, the sample stir-fried with sand being the strongest.It was also firstly confirmed to have the action of analgesia. And the samples stir-fried with sand were more effective than the raw medicinals.The rhizome and hair of C. barometz were firstly found to prolong the clotting time and bleeding time, and the sample stir-fried with sand was the strongest.In the mouse with adjuvant arthritis (AA) and mouse with adjuvant arthritis with deficiency in the kidney (DK-AA), it was confirmed that this medicinal decreased the blood rheology indexes, increased the weight of mouse and their organs' weight, inhibited the turgenscence of the feet, increased the content of trace elements in blood, decreased the content of Interleukin-lp (IL-1) and tumor necrosis factor (TNF-a) in the joint fluid. So it showed the effects of C. barometz on the mouse with AA and mouse with DK-AA. And the sample processed with sand was stronger than the raw one.3. Researches in processing technology and quality standardOn the basis of chemical and pharmacological studies, the best processing technology was optimized by orphonogal test. It would be better to be stir-fried with 8 times of sand at 180 C~190 C for 5 or 6 minutes, thus to provide with theoretical basis for the studies on quality standard of the medicinals.About the quality standard, the processed sample should be controlled with 7-13% of moisture, no less than 0.02% of volatile oil, no less than 0.01% of protocatechualdehyde and protocatechuic acid. 5-hydroxymethyl furfural in the processed sample should be detecte... |
