Recently, there are more and more investigation on IL-1B.NGF and NF-kB aiming at illustrating the mechanism involved in the reduced brain function after sleep deprivation. No such investigation has been found in China so far. Here we use the model of total sleep deprivation of rats and be combined with the approach of immunohistochemistry, Western-blot and RT-PCR to investigate the expression of II-lBand NGF protein in rat's hippocampus and cortex, to investigate and discuss the relation between them and sleep deprivation, to test the change of NF-kB and IkB-a, to investigate and discuss the relation between them and IL-1B and the transformation of NGF. We use naloxone to administer them so that we could investigate and discuss the effect of learning and memory in rat.Parti the effects of total sleep deprivation on IL-1B protein and mRNA expression in rat's hippocampus and cortex.Objectives to investigate the effects of total sleep deprivation on IL-1B protein and mRNA expression in rat's hippocampus and cortex. Methods Put the rats in the rotating drums whose rotating rate were 1/min to make the TSD model in different time course. Divided the 60 rats into total two groups: immunohistochemistry group (n=30) and PT-PCR (n=30), and the two groups should be respectively further divided into 5 small groups randomly: antitheses group(CC group for normal and TC group for environment) and sleep deprivation group ( TSD6h,TSDld,TSD3d (n=6))o we adopt the approach of immunohistochemistry and RT-PCR to test IL-1B protein and IL-1B mRNA expression in rat's hippocampus and cortex. Results the results of immunohistochemistry group showed: IL-1B mostly distributed in cell plasm and the expression of IL-1B was also found in cell nucleus. The cubage of the positive cell was small and its shape was various such as round like an egg, shuttle and triangle, the cell plasm was dense and only have a few prominency, most of them are central nerve cell and some pectin cell; the positive cells mostly distributed in hippocampus CA3 , CA1 and dentation gyrus nucleolus and cell plasm; In cerebral cortex, positive cells distribute in every stratum, and the substance in cell circumference were negative. The results of PT-PCR showed thatthe expression of IL-lBmRNA was significantly higher than antitheses group within the hippocampus area with TSD6h group and TSDld group and also the expression of TSD6h group was significant increased within the CA1 area. Conclusions total sleep deprivation on IL-16 protein and mRNA expression in rat's hippocampus and cortex was higher, and the longer the time course, the more significant the result was. The higher IL-1 8 maybe help to protect the cerebral nerve cell after short-time sleep deprivation, but they maybe harm the nerve cell after overtime in deprivation.Part 2 the effect of total sleep deprivation on NGF protein and mRNA expression in rat's hippocampus and cortex.Objectives to investigate the effect of total sleep deprivation on NGF protein and mRNA expression in rat's hippocampus and cortex. Methods the working approach of grouping for animals, making modal and immunohistochemistry groups and RT-PCR were the same as part one. We use immunohistochemistry to test protein expression in rat's hippocampus and cortex after sleep deprivation and use RT-PCR method to test the expression of NGF mRNA. Results the results of immunohistochemistry group showed that the NGF in TSD group of hippocampus CA1 area was significantly higher than antitheses group but no obviously difference was found. The TSD6h group in CA3 area was significantly lower than antitheses group but the TSD3d group was significantly higher than antitheses group Conclusions The results showed that sleep deprivation can lead to the change of NGF expression in experimental rat's hippocampus and cortex, the short-time sleep deprivation can decrease the expression of NGF but overtime deprivation can increase it. The increased NGF expression can nourish and protect to nerve cell, and repair the injured nerve cell.Part 3 the investigatio...
