Objective: To establish a animal model with neural invasion of adenoid cysticcarcinoma(ACC) and probe to the role of nerve growth factor(NGF) in neural invasionof ACC.Methods: ①In order to establish a neural invasion model of ACC in animal, 5×105ACC cells or tongue cancer cells(for control) were subcutaneously injected intoinfraorbital points in nude mice and neural invasions in infraorbital nervi were observedusing H&E staining. ②Expression of NGF was detected in ACC cells in vitro byWestern Blotting and that in vivo by immunohistochemistry. ③Changes of neuralinvasion were observed in nude mice with intra-abdominal injection of anti-NGFantibody. Effect of anti-NGF antibody on ACC cell proliferation was evaluated by MTT.Results: ①Animal model with neural invasion of ACC was successfully establishedin nude mouse. Neural invasions in infraorbital nervi were produced, and incidence ofneural invasions of ACC is higher than that of tongue cancer in this model. ②Cells ofACC in vivo or in vitro presented NGF, and expression of NGF in ACC adjacent tonervi was significantly higher than that far from nervi. ③Anti-NGF antibody markedlysuppressed the neural invasion of ACC in vivo and reduced ACC proliferation in vitroin a dose-dependent manner.Conclusion: This animal model is a good model for study neural invasion of ACC.Enhanced expression of the NGF in ACC was closely correlated with neural invasion.Mechanisms of NGF promoting to the neural invasion in ACC involved thechemoattraction between NGF in ACC and nerve growth factor receptor(NGFR) innerve and enhancement of ACC cell proliferation by NGF in a dose-dependent minner.
