Continuous research effort headed by one of the pioneers of organ preservation, Folkert O. Belzer, resulted in a development of a new preservation solution that introduces three new philosophies in 1988 : (a) osmotic concentration is nolonger maintained by metabolically active glucose, it is rather achieved by the administration of metabolically inert substrates like lactobinat and raffinose; (b) much attention is paid to the additional administration of colloid carrier hydroxyethylstarch (HES); (c) addition of oxygen radical scavengers, glutathion, allopurinol, and adenosin.This solution has proven to guarantee extreme long cold ischemia tolerance, 72 hours in kidneys6 and certainly 487 hours in livers. This solution was also tested prospectively in a multicenter European trial, where University of Wisconsin solution (UW) has been found superior to Eurocollins by decreasing the rate of delayed graft function from 33% to 23%. At the moment, UW is considered the standard preservation solution for livers, kidneys, and pancreases. Attempts to use UW for heart preservation were not convincing.Belzer solution (UW) made great progress in preservation, at the same time, made great progress in price, about 220 $ /L. That high price is a burden to the society even in developed countries, so, much attention have been paid to inventing a cheap substitute with same effect. Because the cost of inventing a whole new solution is too high to pay, We made a selection to improve HC-A solution that has been widely employed and proved to be satisfied in renal preservation, the primary outcome of improvement was named F-Z solution.We compared the preservation effect of the three kinds of solution in cellularlevel at 4#. We compared LDH concentration in superior schedual of cultivated endothelial cell (ECV/304 cell line) that been preservated and rewarwed to reflect I-R injury, compared metabolism of MTT after rewarming to reflect the vital force.After two years hard work, we achieved some conclusion in cellular level: 1. Possibility to improve HC-A solution is being; 2. As the primary'outcome of improvement, F-Z solution is better than HC-A solution, arrives the similar level of Belzer solution in decreasing I-R injury and improving the vital force after preserve-rewarmimg; 3. I-R injury can completely present only after rewarming, more serous the sustainable I-R injury is, more quickly it presents after rewarming. 4. The vital force after preserve-rewarmimg presents fluctuation and periodicity, during every period there can be a "best preserve-rewarmimg time (range)" to increase the vital force after preserve-rewarmimg in theory.In the second part of this paper, we explored the principle of cryopretectants that still unclear after being widely employed more than half century by a very simple-experiment. On the base of the experiment's outcome, we put forward a hypothesis: The principle of cryopretectants is that cryopretectants can put down the solution's freezing point, equal to even under intracellular fluid's, to make it possible that a full round ice layer inside preservated cell membrane comes to being at the beginning of the freezing procedure. On the basis of the hypothesis, we perfectly answered several hard traditional theoretical questions, and criticized vitrificaton.
