Herba Ephedrae Decoction (HED) is composed of Herba Ephedrae, Ramulus Cinnamomi, Semen Armeniacae Amarum and Radix Glycyrrhizae. The compatibility of HED is very right-sizing, besides the traditional Chinese doctors from the past dynasties have the identical understanding about the HED. So we think HED has the better representational to study the compatibility principles of prescriptions of Traditional Chinese Medicine.Objective: To analyze the function and status in HED of Ramulus Cinnamomi, Semen Armeniacae Amarum and Radix Glycyrrhiza, and further to discuss the compatibility rule of HED, by the change of dissolvable quantity of ephedrine(E) and pseudoephedrine(PE) in each decoction with Herba Ephedrae, the pharmacokinetics of E and PE in the different viscera tissue, the concentration of amino acids neural transmitter in prefrontal cortex of the rats .Methods: Fixed the Herba Ephedrae, Ramulus Cinnamomi, Semen Armeniacae Amarum and Radix Glycyrrhizae were acted as the three factors , "yes" and "no" as the two levels of each factor, used the L8(27) orthogonal design table to carry out the grouping, gained eight groups with Herba Ephedrae. Upon the above groundwork, we completed the experimentations, as follows:(1) Used the E and PE as analytical index, the trifluoroacetic anhydride asthe derivatizing agent, GC/MS/SIM, researched the change of dissolvable quantity of E and PE in each decoction with Herba Ephedrae, and determined the concentration-time value of E and PE in the different viscera tissue, and utilized the noncompartmental model in the PK software-WinNonLin program to compute the PK parameters, but AUC was calculated by the trapezia-area means, the Cmax Tmax were calculated by the three-point parabolas. Whereafter, the main parameters, AUC, AUC/D,Cmax, Cmax/D,Tmax,t1/2, MRT and CL were analyzed by the General linear model-univariate in SPSS 10.0.(2)Used 2,4-dinitrofIuorobenzene as derivatizing agent, HPLC, determined the content of Glutamic acid, Aspartic acid, y-aminobutyric and Glycin in prefrontal cortex of the rats. First, researched the dose-response and time-response relationship of HED, and ascertained the best dose and time. Upon the groundwork, analyzed the effect of compatibility on the content of the amino acids, while set up the group without Herba Ephedrae and solvent group as control group, the data were statistically analyzed by Independent- Samples T test and the General linear model-univariate in SPSS 10.0, respectively.ResuIts: (1) Ramulus Cinnamomi had significantly reduced the dissolvable quantity of E and PE in the decoctions(P<0.05), Semen Armeniacae Amarum had significantly reduced the dissolvable quantity of PE(P<0.05), and had not significant effect on E(P<0.05), while Radix Glycyrrhizae and the all two-way interactions had not significant effect on E and PE (P<0.05).Ramulus Cinnamomi had significantly reduced the AUC and AUC/D value of E in the brain(P<0.05), but had not significant effect on the AUC and AUC/D value of E in the lung, heart, liver and kidney; had not significant effect on the Tmax Cmax Cmax/D t1/2 MRT CL value of E in all analyzed tissues(P>0.05). However, the effect of Ramulus Cinnamomi on all parameters of PE in all analyzed tissue was not significant (P>0.05).Semen Armeniacae Amarum had significantly reduced the AUC and AUC/D value of E in the brain and liver(P<0.05 ), and had not significant effect on other tissue and not significant effect on PE in all tissue (P>0.05). Semen ArmeniacaeAmarum had significantly reduced the T value of E and PE in the liver tissue, significantly increased the C /D value of PE in the kidney(P<0.05 ), not significant effect on C/D value in other tissue and t1/2% MRT\ CL value in all analyzed tissues(P>0.05).Radix Glycyrrhizae had significantly increased the AUC/D value of E in the brain(P<0.05 ), but reduced the AUC and AUC/D value of E in the liver(P<0.05 ), not significant effect on AUC and AUC/D value of PE in all analyzed tissue and not significant effect on Tmax Cmax Cmax/TX t1/2 MRT CL value in all...
