Objective: Probe into the mechanism of regulation of TaiYang and YinTang electro- acupuncture on the sleep of rats. Methods: 200 wistar rats were selected into 4 parts of the research. Duplicate insomnia model on rats by means of PCPA abdominal injection. Take TaiYang and YinTang electro-acupuncture as therapy method while Diazepam administration were taken by control group. Select the change of EEG and content of 5-HT, 5-HIAA, NE, IL-1, TNF, GABA and immunohistochemical location of GABAA receptor in the brain of rats as observing standards. Results: 1 .Acupuncture therapy prolonged the sleeping time of rats coursed by sodium pentobarbital in which slow wave sleep were increased most with significance of P<0.01. 2. Both of acupuncture and Diazepam returned the sleep of insomnia rats caused by PCPA. 3. After PCPA abdominal injection the normal sleep of rats disappeared, at the same time, the content of 5-HT and 5-HIAA and immunohistochemical location of GABAA receptor in the brain of rats decreased with comparison to normal group(P<0.05,P<0.01), but the content of NE, IL-1, TNF and GABA did not change much compared with normal group. 4. After Diazepam administration, the content of 5-HT, 5-HIAA, NE, IL-1 and TNF in the brain of insomnia rats had no obvious change while that of GABA increased comparing with normal group(P<0.01) and immunohistochemical location of GABAA receptor increased comparing with model group(PO.Ol). 5. After acupuncture treatment the content of 5-HT and 5-HIAA increased in contrast with model group(P<0.05,P<0.01) while that of NE decreased in contrast with normal group(P<0.05). At the same time, the content of IL-1, TNF and GABA increased greatly with comparison to normal group(P<0.05, P<0.01), the immunohistochemical location of GABAA receptor also increased greatly with comparison to model group (P<0.01). Conclusion: Acupuncture is superior to Diazepam in the treatment of insomnia due to its good regulation on slow wave sleep, neurotrasmitters and immune cytokine.In general, acupuncture therapy is an effective method in the treatment of insomnia. Furthermore, it has good adjustment on nervous and immune system in the body of the insomnia. This will afford objective theoretical evidence to the clinical treatment of insomnia.
